摘  要:目的研究性激素对MRL/lpr狼疮模型鼠Th1/Th2类细胞因子表达的影响,探讨其对Th1/Th2平衡的调节作用。方法将狼疮模型鼠去势后随机分为空白对照组、雌二醇(E2)生理水平组、雌二醇(E2)妊娠水平组、睾酮(T)组。利用RT—PCR方法检测模型动物血液单个核细胞中Th1类细胞因子(IFN-γ,IL-2)、Th2类细胞因子(IL-4,IL-6)的基因转录状态。结果雌二醇生理水平组和妊娠水平组IL-2 mRNA的水平低于空白对照组(P〈0.05),雌二醇妊娠水平组IL-4和IL-6的mRNA水平高于空白对照组(P〈0.05)。睾酮组IL-6mRNA州平低于空白对照组(P〈0.05)。结论性激素可以调节Th1/Th2细胞因子的表达水平,妊娠剂量的雌激素使狼疮模型剥的细胞因子向Th2偏移,而雄激素则抑制Th2偏移。Objective To study the influences of sexual hormones on the expression of Th1/Th2 cytokines in MRL/lpr lupus model mice and investigate it's modulation on Thl/Th2 balance. Methods The ovariectomized lupus model mice were divided into blank control group,estradiol (E2) group of physiologic level ,estradiol(E2) group of gestational level,testosterone(T) group. Thl cytokines (IFN-Y, IL-2) and Th2 cytokines (IL-4,IL-6) in peripheral blood mononuclear cell (PBMC) from the animal model were detected by RT-PCR. Results The level of IL-2 mRNA in estradiol(E2) group of physiologic level and gestational level were significantly lower than that of blank control group(P〈0. 05). The level of IL- 4 and IL-6 mRNA in estradiol(E2) group of gestational level were significantly higher than that of blank control group(P〈0. 05). The level of IL-6 mRNA in testosterone(T) group was significantly lower than that of blank control group(P〈0. 05). Conclusion Sexual hormones can modulate the expression levels of Th1/Th2 cytokines. Estradiol of gestational dosage can make the cytokines of lupus model mice Th2 excursion,however androgen can repress it.
