酵母样真菌液体培养药敏试剂盒的研制

摘　　要：目的为酵母样真菌药敏提供操作简便、经济实用、准确可靠的体外液体药敏试剂盒,给临床用药提供参考依据。方法参照美国国家临床实验室标准化委员会(NCCLS)M27-A方案,根据药物MIC值,参照文献建立高低两个药敏浓度的液体培养药敏试剂盒;用标准参考菌株(c.p,ATCC22019、c.k,ATCC6258)为试验菌株测定试剂盒的重复性和稳定性;用221株临床分离株与ATB FUNGUS3药敏试剂盒进行比较。结果批内MIC符合率为97.50%,批间MIC符合率为95.83%;试剂盒在-20℃保存6个月内稳定,MIC符合率都在90.00%以上;与ATB FUNGUS3药敏试剂盒一致性较好,Kappa>0.7,P<0.05。结论酵母样真菌培养药敏试剂盒可以满足临床检验需求,值得推广应用。Objective To find one kind of reagent kit with single operation,economical practication and liable accuracy, and to bring a referance for using some clinical drug. Methods Refering to national committee for clinical laboratory standards （ NCCLS） M27-Ascheme, developed according to the lower and the higher density liquid culture kit for yeast-like fungi drug sensitivity test was the drug MIC reference book. The repetition and the stabilization of the reagent kit was determined by the standarding bacteria frunk （C. parapsilosis ATCC 22019 ,C. krusei ATCC6258） as experimental bacteria trunk. Then the 221 trunks of Clinical bacteria were compared with that of ATB FUNGUS 3. Results There was 97.50% to accord with the MIC in the same kits, but it was 95.83% on the different kits ;The reagent kits incubated at -20℃ were stable during 6 months, it was above 90.00% to accord with the MIC. The reagent kits were consistent with ATB FUNGUS 3, Kappa 〉 0.7, P 〈 0.05. Conclusion The liquid culture kit for Yeast-like fungi drug sensitivity tests can be satisfied with clinical examination and maybe applied spreadly.
