摘  要:目的建立测定小鼠血浆、心、肝、脾、肺、肾、脑中醋酸地塞米松含量的方法。方法色谱柱为Shimpack C18柱,流动相为甲醇-水(65:35),流速为1ml·min^-1,内标为甲睾酮,检测波长为240nm。结果在生物样品中,醋酸地塞米松浓度1~100ug·g^-1与峰面积比呈良好的线性关系,最低检测限为40ng·g^-1;方法回收率和RSD值均符合生物样品测定的要求。结论所建方法的线性、精密度、回收率均良好,且简便灵敏,可用于醋酸地塞米松制剂的药物动力学研究。OBJECTIVE To establish a method for the determination of Dexamethasone acetate in rat plasma, heart, liver, spleen, lung, kidney and brain. METHODS Dexamethasone acetate was separated from Shimpack C18 column with methyhestosterone as the internal standard. The mobile phase consisted of methanol -water(65 : 35) at a flow rate of 1 ml .min^-1. The UV detection wavelength was set at 240 nm. RESULTS The linear range of Dexamethasone acetate in biolgogical specimen was 1 - 100 ug.g^-1. The lowest detectable limit was 40 ng.g^-1. The recovery and RSD of the method met the requirements for bioassay. CONCLUSION With good linearity, precision and recovery, the method is sensitive and simple, and suitable for pharmaeokinetie study of Dexamethasone acetate preparation.
