摘 要:本实验对体外培养的新生大鼠心房和心室肌细胞进行了光镜和电镜观察,并以免疫组织化学方法对心肌细胞内心钠素的定位和含量进行了探讨。结果表明,培养的大鼠心房和心室肌细胞胞浆内均可见到心钠素免疫反应阳性颗粒,但心室肌细胞内心钠素含量很少;并观察到有含和不含心钠素阳性颗粒的两种心房肌细胞。实验证实,心钠素在培养新生大鼠心肌细胞中的定位与含量与在体时基本一致。本方法为从细胞水平研究各种因素对心钠素释放的影响提供了新途径。The morphology of cultured atrial and ventricular myocytes from newborn rats was studied under light microscope and electron microscope, and the localization of the atrial natriuretic factor (ANF) was carried out by means of immunohistochemistry PAP method. The results showed that the ANF immunoreactive substance was found in both the atrial and ventricular myocytes, but there were fewer in the ventricular myocytes than the atrial myocytes. The atrial myocytes might be classified into two kinds, namely the ANF positive and negative cells. The results revealed that the localization of ANF in culture cardiomyocytes from the neonatal rats was identical to that demonstrated in vivo.
