摘 要:本实验选用体重220~280g的雄性Wistar大鼠20只。取其左侧跖肌组织块做恒冷箱连续切片。将相邻切片做不同的组织化学反应,包括肌球蛋白ATPase、肌糖原、肌脂类、琥珀酸脱氢酶、细胞色素氧化酶、乳酸脱氢酶、6-磷酸果糖激酶及非特异性酯酶。用显微分光光度计对肌纤维的各种组织化学反应进行了定量测定。结果发现,根据大鼠跖肌肌球蛋白ATPase反应(碱性预孵育,pH10.4及酸性预孵育,pH4.65)可将肌纤维分为Ⅰ型、ⅡA型和ⅡB型3种纤维。各型纤维的其他组织化学反应不尽相同。糖原、乳酸脱氢酶、6-磷酸果糖激酶在Ⅱ型纤维分布较多;脂类、琥珀酸脱氢酶、细胞色素氧化酶在Ⅰ型和ⅡA型反应较强、反应强度的顺序为ⅡA>Ⅰ>ⅡB。非特异性酯酶的强度顺序为Ⅰ>ⅡA>ⅡB。所得结果说明,大鼠跖肌不同类型肌纤维存在着明显的代谢差异。其中ⅡA型纤维既具有很高的氧化活性,又具有很高的糖酵解活性,在骨骼肌能量代谢方面可能具有重要意义,值得深入研究。Twenty male Wistar rats (220-280 g in body weight)were used. Left plantaris were removed and serial eryostat sections were made immediately. Eight histochemical methods (including lipid, glycogen, m-ATPase, SDH, CCO, LDH, PFK, and ANAE) were used to identify the properties of skeletal muscle fibers. The histochemical reactions of different fibers were quantitated by microspectrophotometer. This experiment showed that three fiber types (Ⅰ, ⅡA and ⅡB) were distingushed based on the myofibrillar actomyosin ATPase during acid preincubation (pH 4.65) and alkaline preincubation (pH 10.4). The quantitative results demonstrated that the activity of lipid, SDH and CCO reactions was strong in type ⅡA fibers, the sequence of three type fibers was ⅡA>Ⅰ>ⅡB, whereas the activity of ANAE reaction was strong in type Ⅰ fibers, the sequence was Ⅰ>ⅡA>ⅡB. Our investigation also showed that the activity of PAS, LDH and PFK was stronger in type Ⅱ fibers than in type Ⅰ. It was concluded that each type fibers in the rat plantaris exhibit significant differences in metabolic and functional activity. The type ⅡA fibers present both high activity of glycolytie enzyme and oxidase, and reflect that these enzymes may play very important role in energy metabolism. Further study remains to bedertaken.
