摘 要:目的比较RA(retinoid acid)和cAMP诱导神经干细胞表达离子通道亚型的区别。方法用RA和cAMP两种方法体外诱导神经干细胞成为神经元,然后用RT-PCR和定量PCR方法对离子通道亚型进行筛选;用膜片钳记录Na+离子电流;用共聚焦记录Ca2+内流。结果RA可以显著上调L-Ca2+通道表达;而cAMP则上调Na+离子通道β-亚基(SCN1B、SCN2B、SCN3B)的表达。cAMP诱导的神经细胞可以记录到特征性的Na+内流而RA没有;RA诱导的神经细胞对神经递质Ach有较强的Ca2+内流应答,而cAMP较弱。结论RA和cAMP诱导神经干细胞表达不同的离子通道亚型。Objective To compare the different subtypes of ion channels expressed in neural progenitors after treatment with retinoid acid (RA) and cAMP. Methods Neural progenitor ( hNPC - TERT cell line) cells were treated by RA or cAMP. Ion channel subtypes were detected by RT - PCR and real time PCR. The inflow current through sodium channels were tested by patch clamp, while the calcium inflow through calcium channels were conformed by Fluo - 3 [ Ca^2+ ] method. Results L - Ca^2+ channels were up regulated in neural progenitors treated with RA. And these cells could respond to Ach (acetylcholine). Sodium channel expression was up regulated by cAMP treatment, especially the β -subtypes (SCN1B, SCN2B and SCN3B). Sodium currents were recorded by patch clamp in these cells. Conclusion RA and cAMP may have different effects on the expression of sodium and calcium channels.
