摘 要:采用农杆菌介导法对新疆甜瓜进行了遗传转化研究,对影响转化频率的不同因素,即:抗生素浓度、预培养有无及感染时菌液的不同处理、外植体与农杆菌的感染时间和共培养时间等进行了研究,通过农杆菌介导法将雪花莲凝集素基因导入四日龄甜瓜子叶外植体,获得了卡那霉素的抗性植株。移栽成活并生长良好。卡那霉素抗性鉴定及转化株总DNA的PCR分析表明,GNA基因已整合到甜瓜的基因组中。初步抗蚜虫实验证明,转基因甜瓜有一定的抗棉蚜幼虫效果。This study was designed to investigate the genetic transformation of Cucumis melo L. by Agrobacterium- mediated trans formation. Some factors affecting the Agrobacterium - mediated transformation frequency, i. e. concentrations of antibiotics, preculture of cotyledon explants, treatments of Agrobacterium and infection time, co-infection time were also discussed. GNA Gene was translated into melon by Agrobacterium - mediated transformation , The kanamycin-resistant plants have been regenerated. They survived after transplantation and grew well in pots. Transgenic plants were confirmed by the kanamycin- resistant experimen and PCR analysis. The preliminary results from bioassay demonstrated significant resistance of the transgenic plants to cotton aphide larvae.
