摘 要:目的:以供体造血干细胞(HSC)诱导异基因大鼠肾脏移植免疫耐受,探讨免疫耐受的形成机制。方法:建立大鼠肾脏移植模型,通过免疫磁珠法负筛选系统提取供者骨髓中的造血干细胞。给低剂量放射线全身照射的Wistar大鼠管饲CsA,接着经尾静脉推注SD大鼠的造血干细胞,并当天完成肾移植。观察各组大鼠的存活时间,监测血清肌酐浓度,并进行移植肾彩色多普勒超声。采用体外混合淋巴细胞反应(MLR)法检测长期存活大鼠的免疫耐受状态。结果:实验组大鼠存活时间与对照组比较有显著差异。混合淋巴细胞培养提示,长期存活的受者脾细胞对供者脾细胞的刺激不表现出明显细胞增殖反应,而对无关大鼠脾细胞产生较明显的增殖反应。结论:应用造血干细胞能够成功地诱导出针对供者特异性的免疫耐受。Objective: To induce immune tolerance to allogenic kidney transplantation with hematopoietic stem cells (HSC) in recipient rats and to study its mechanism. Methods: A model of kidney transplantation was established. HSC were collected by StemSepTM. Wistar rats were treated with TBI and fed with CsA. HBC were injected into recipient rats and kidney transplantation was performed on the same day. The survival time of recipient rats was observed. Serum creatinine was measured and color Doppler flow imaging was checked. The immunologic tolerance status in rat renal allografts was detected by MLR on day 30, the recipient rat spleen cells were cuhured with donor spleen cells as a third part. Results: The difference was significant between the case and other three control groups. Hypo-response to the donor occurred while the normal reaction to the third parity was retained. Conclusion: Hematopoietic stem cells can induce donor-specific tolerance to allografts.
