摘  要:为观察出血性休克后复苏开始前给予阿片δ1受体激动剂2-Methyl-4aa-(3-hydroxyphenyl)-1,2,3,4,4a,5,12,12aα-oc-tahydroquinolino[2,3,3-g]isoquinoline dihydrobromide(TAN—67)对大鼠心肌缺血缺氧性损伤的影响。将24只成年雄性SD大鼠,随机分为三组,每组8只,实验分假手术组、休克对照组、TAN-67组。休克对照组和TAN-67组大鼠放血使MAP降至40mmHg,休克持续60min,注射生理盐水或TAN-67后,回输自体血进行复苏,观察复苏2h内MAP和心功能(LVP、±dp/dtmax)的变化。实验结束时取心肌组织行病理学电镜检查。并分析各组线粒体损伤。休克对照组复苏后各时点血流动力学指标呈逐渐下降趋势。TAN-67组复苏后同一时点各项指标均显著高于对照组(P<0.05)。病理学电镜检查和线粒体损伤结果,TAN-67组心肌损伤较对照组减轻。结果表明TAN-67有利于急性出血性休克大鼠血流动力学复苏,对心肌损伤有保护作用。To investigate effects of δ1 opioid receptor agonist 2-Methyl-4aα-(3-hydroxyphenyl) -1,2,3,4,4a,5, 12,12aα-octahydroquinolino [ 2,3,3-g ] isoquinoline dihydrobromide ( TAN-67 ) on myocardial hypoxic-ischemic injury in rats with hemorrhagic shock, twenty-four male SD rats were randomly divided into 3 groups (n = 8 each) : sham group,control group and TAN-67 group. The animals in control and TAN-67 groups were exsanguinated till MAP decreased to 40 mmHg. Rats were resuscitated with shed blood after administered to normal saline or TAN-67 at the end of shock period. Mean arterial pressure (MAP), cardiac function (LVP, + dp/dtmax) were recorded after resuscitation for 2 h. Morphological changes in myocardium were observed by electron microscope. FlaMeng' semi-quantitative analysis of mitochondria was used to analyze electron microscope specimens. Higher scores indicated more serious damage. After hemorrhagic shock and resuscitation, hemodynamics gradually decreased in control group. While hemodynamics in TAN-67 group are significantly increased compared with control group. Myocardial injury is less severe than that in control group. TAN-67 could improve the hemodynamics and reduce myocardial injury during the course of hemorrhagic shock.
