颐脑解郁方对出血性脑卒中后抑郁大鼠额叶皮质及海马PDE_4 mRNA的调节作用

摘　　要：目的:观察出血性脑卒中后抑郁模型大鼠脑内PDE4mRNA的表达及中药颐脑解郁方的干预作用。方法:采用逆转录PCR(RT-PCR)方法,对PDE4mRNA在出血性脑卒中后抑郁大鼠脑内前额叶皮质、海马的表达及中药颐脑解郁方的干预作用进行观察。结果:出血性脑卒中后抑郁模型大鼠脑内前额叶皮质、海马区域PDE4mRNA的表达明显高于正常组(P<0.01),中药颐脑解郁方能明显降低PDE4mRNA的表达(P<0.01)。结论:出血性脑卒中后抑郁模型大鼠脑内PDE4mRNA的表达增强,中药颐脑解郁方能使之明显降低,提示出血性脑卒中后抑郁症与脑内PDE4mRNA表达增强有关,抑制PDE4的表达是颐脑解郁方抗抑郁机制之一。Objective ： To observe the expression of PDE4 and the interference effect of Yinaojieyu formula on the rat model of post - stroke depression. Methods ： Reverse transcription polymerase chain reaction （ RT - PCR） was used to determine the expression of PDE4 mRNA in prefrontal cortex and hippocampus, and the interference of Yinaojieyu formula was observed. Results：Compared with normal group,the expression of PDE4 mRNA in hippoeampus and prefrontal cortex in model group increased obviously （ P 〈 0.01 ）. Compared with the model group ,the expression of PDE4 mRNA in Yinao- jieyu formula group decreased significandy （ P 〈 0.01 ）. Conclusion ： The expression of PDE4 mRNA in the brain of rat model of post - stroke depression increased,and Yinaojieyu formula could decrease this expression. The anti - depression effect of Yinaojieyu formula may be related to its function of decreasing PDE4 mRNA expression.
