益气养阴活血化瘀法对肺纤维化鼠模型的干预作用及对TGF-β1表达的影响  被引量:4

摘  要:目的:通过观察丹参、参麦注射液对博莱霉素致肺纤维化模型大鼠肺组织病理学、免疫组化肺组织中TGF-β1表达和支气管肺泡灌洗液中TGF-β1表达的定量分析,明确益气养阴、活血化瘀法对肺间质纤维化的治疗作用并探讨其作用机理。方法:采用BLMA5致大鼠肺间质纤维化的造模方法,随机将72只大鼠分为4组:空白组、博莱霉素模型组、丹参组、丹参合参麦组,除空白组外其余3组分别每天腹腔注射生理盐水、丹参注射液、丹参合参麦注射液,各组分别于第7、14、28天各处死6只大鼠,依次收集支气管肺泡灌洗液,取肺病理标本。通过计算机图像分析系统定量分析第28d的病理改变,免疫组化图像分析测量第7、14、28天的TGF-β1吸光度,以及ELISA酶法测定支气管肺泡灌洗液中第7、14、28天的TGF-β1含量。结果:经丹参和丹参合参麦注射液干预后,大鼠的纤维化和肺泡炎均较模型组轻,丹参合参麦组比丹参组更轻(P<0.05)。丹参组和丹参合参麦组的肺组织免疫组化TGF-β1平均吸光度、肺泡灌洗液的TGF-β1的含量均较模型组低(P<0.01),其中第14天丹参合参麦组较丹参组更低(P<0.05)。结论:以丹参合参麦为代表的益气养阴、活血化瘀法对肺纤维化有明显改善作用,其作用机理与其降低TGF-β1的表达有关。Objective : To observe the effect on the rat models of pulmonary fibrosis by Danshen and Shenmai parenteral solution with the pulmonary pathological changes, expression of TGF - yiqiyangyin; huoxuehuayu; bleomycin; interstitial pulmonary fibrosis ; TGF - β1 in pulmonary tissue and bronchoalveolar layage fluid (BALF). And discuss the action mechanism of yiqiyangyin and huoxuehuayu methods in the pulmonary fibrosis. Methods: Reproduce the rat models of pulmonary fibrosis by injecting bleomycin to trachea,72 SD rats were randomly divided into four groups, including the normal group, the modeled group , the Danshen group, the Danshen and Shenmai group. Then treat the latter three rat models with intraperitoneal injection by physiological saline, Danshen parenteral solution, Danshen and Shenmai parenteral solution meanwhile. The rats in each group were partially killed in 7th, 14th, 28th day, the BALF was selected and the pulmonary tissue was incised to make pathological sections which were stained with HE and were stained with TGF -β1 immunohistochemical techniqu. Using computer image analysis system evaluate the result of the pulmonary pathological changes of each group on the 28th day. Also the expression of TGF - β1 in pulmonary tissue was analyzed with comptuer image system and the concentration of TGF - β1 - protein in BALF was detected by way of ELISA on 7th, 14th,28th day. Results: Treated with Danshen parenteral solution, Danshen and Shenmai parenteral solution, the pulmonary fibrosis and pulmonary alveolitis of rat models could be alleviated, and the latter group could be significantly decreased( P 〈0.05). At the same time, we also observed that in the former two groups the expression of TGF - β1 in pulmonary tissue and BALF was lower than the modeled group( P 〈 0.01 ), and on the 14th day the Danshen and Shenmai group was lower than the Danshen group( P 〈0.05).
