摘 要:目的:探讨耳针联合阿尼西坦(Ani)治疗血管性痴呆(VD)的中枢机制。方法:将60只VD模型大鼠分为四组:模型组、耳针组、阿尼西坦组(Ani组)、耳针及阿尼西坦联合组(联合组),每组15只;另选15只正常大鼠作为对照。用Morris水迷宫实验检测大鼠学习记忆能力;记录大鼠海马齿状回长时程增强(LTP)的电生理指标。结果:①对照组、耳针组、Ani组和联合组大鼠的学习记忆能力均优于模型组,P〈0.05或P〈0.01;联合组与耳针组、Ani组比较,在部分时段有差异,P〈0.05。②对照组、耳针组、Ani组及联合组与模型组比较,海马齿状回LTP的维持和诱导明显优于模型组,P〈0.05或P〈0.01;联合组与耳针组和Ani组比较,在部分时段有差异(P〈0.05)。结论:耳针及阿尼西坦能改善VD大鼠学习记忆能力和海马LTP的维持和诱导,其机制可能是通过改善海马神经突触的可塑性而实现的。Objective: To investigate the mechanism of ear acupuncture and aniracetam (Ani) on vascular dementia (VD)model rats in vivo. Methods: Sixty model rats with VD were divided into 4 groups,the model group, the ear acupuncture group, Ani group and the combined group (ear acupuncture and Ani), Other fifteen rats were controls. The capabilities of learning and memory of the experimental animals were assessed through Morris Water Maze Behavior Test, Long term potential (LTP) was recorded in the hippocampal dentate gyrus as electrophysiologic index for evaluation of learning and memory. Results: (1) Compared with the rats in the model group, the learning and memory abilities of controls, the ear acupuncture ear group, Ani group and the combined group were more effective, P〈0. 05 or P〈 0.01. Compared with the rats in the ear acupuncture group anti Ani group, the combined group was the most effective in part time,P〈0.05. (2) Compared with the rats model group , the inducement and maintenance of LTP in hippocampal dentate gyrus of controls,in the ear acupuncture group, Ani group and the combined group were more effective, P〈0.05 or P〈0.01. Compared with the rats in the acupuncture group and Ani group, and the combined group was the most effective. P〈0. 05. Conclusion: Both acupuncture ear and Ani could facilitate the inducement and maintenance of LTP in hippocampal dentate gyms,the combined group was more effective and the mechanism were realized possibly by improving the synaptic plasticity in hippocampus.
