摘 要:目的:观察鞘内注射奈福泮及可乐定对福尔马林致痛大鼠的镇痛效应及对脊髓背角c-fos表达的影响。方法:选择鞘内成功置管的雄性SD大鼠32只,随机分为4组(n=8):对照组(S)鞘内注射生理盐水20μL,奈福泮(Nfp)组鞘内注射20μg奈福泮,可乐定(C)组鞘内注射20μg可乐定,奈福泮和可乐定联合用药(Nfp+C)组鞘内注射奈福泮10μg和可乐定10μg。在大鼠足底部注射5%福尔马林后立即记录1 h内每5 min的缩腿、舔爪时间。足底注射福尔马林后2 h将所有动物处死取脊髓膨大处,采用免疫组织化学方法观察脊髓背角c-fos的表达。结果:各组大鼠第二相缩腿、舔爪累计时间进行比较,联合注射组的缩腿、舔爪累计时间显著短于其他组(P<0.01);联合注射组大鼠的脊髓背角FLI-N(c-fos免疫阳性反应神经元)数目明显低于单独用药组(P<0.01)。结论:奈福泮与可乐定鞘内联合应用可能具有协同镇痛作用。Objective:To observe the analgesic effect of intrathecal Nefopam Hydrochloride and Clonidine on the rats induced pain by Formalin and the c-los expression in dorsal horn of spinal cord. Methods: Thirty-two male SD rats were randomly divided into 4 groups( n -- 8 each). They were a control group, which was injected normal saline 20 μL in- trathecally, a Nefopam Hydrochloride (Nfp) group, which was injected Nefopam Hydrochloride 20 μg intrathecally, a Clonidine group(C), which was injected Clonidine 20 μg intrathecally and a Nefopam Hydrochloride combined with Clonidine group(Nfp + C), which was injected Nefopam Hydrochloride 10μg and Clonidine 10μg intrathecally. The time of drawing legs and licking claws was immediately recorded every 5 minutes within an hour after 5 % formalin was injected into planta. All the animals were killed to take some tissue in the enlarged lumbar spinal cord after 2 hours of injecting 5% formalin into planta to observe the expression of c-fos in dorsal horn of spinal cord by immunohistochemistry. Results:Compared the accumulative time of drawing legs and licking claws of phase Ⅱ in rats of each group respectively, the accumulative time of drawing legs and licking claws of combined group was obviously shorter than that of other groups (P 〈0.01). Numbers of FLI-N( C-Fos like immunoreactive neurons) of the spinal cord in the combined group were significantly lower than those of single-drug group ( P 〈 0.01 ). Conclusion: The combination of Nefopam Hydrochloride with Clonidine may have obvious synergistic analgesic effect.
