摘 要:目的:对基因座D1S1676的基因多态性和法医学意义进行研究。方法:山西汉族人群中随机筛选110例无关个体,提取静脉血,用GDB中的引物进行PCR扩增,银染显色。对其筛选的等位基因测序,构建等位基因分型标准物;按照ISFG原则命名各等位基因。结果:本研究首次选择了常染色体基因座D1S1676,该基因座为4核苷酸简单重复STR,重复单位为[GAAG]n。在110例山西汉族无关个体血样中共检出了6个等位基因。其个人识别能力(DP)和非父排除率(PE)为0.920和0.630,其基因型分布符合Hardy-Weiberg平衡(P>0.05)。结论:基因座D1S1676具有较高的遗传多态性,在法医学及人类遗传学方面具有较高应用价值。Objective:To study genetic polymorphism of DIS1676 locus and its forensic significance in Shanxi Han population. Methods:One hundred and ten unrelated individuals were randomly selected to have intravenous blood DNA extracted from peripheral blood and then carried out PCR amplification by primer in GDB and visualized by silver stai- ning. The alleles screened were sequenced to establish standard allelic ladder of DIS1676 locus and then to name each alleles according to the principle of ISFG. Results: An autosomal locus D1S1676 had been first chosen in the study, which was a simple tetranucleotide STR and the repeat unit was [ GAAG ] n. Six alleles had been detected from blood samples of l l0 unrelated individuals. Both of the discrimination power(DP) and the parentage exclusion probability (PE) were 0. 920 and 0. 630. The genotype distribution of DIS1676 locus was in accordance with Hardy-Weinberg equilibreum (P 〉0.05 ). Conclusion :The locus D1S1676 has higher genetic polymorphism and application value in forensic medicine and human genetics research.
