摘 要:目的观察加味逍遥散对慢性应激小鼠学习记忆及免疫细胞的影响,为探讨加味逍遥散的药理作用,提供临床实验依据。方法BALB/c小鼠随机分为5组:正常对照组、应激组、逍遥散组、加味逍遥散低剂量组、加味逍遥散高剂量组。通过水迷宫实验观察小鼠学习记忆能力。采用流式细胞仪检测小鼠脾组织中CD4、CD8的含量。结果加味逍遥散高、低剂量组、逍遥散组与应激组相比均能缩短逃避潜伏期(P<0.01)、增加第一象限停留时间百分比(P<0.01)。小鼠脾淋巴细胞悬液中CD4+T细胞高于应激组(P<0.05),而CD8+T细胞低于应激组(P<0.05)及CD4/CD8比值低于应激组(P<0.05)。结论逍遥散和加味逍遥散均具有显著拮抗因应激所致的学习记忆能力和免疫功能的改变,以高剂量加味逍遥散作用最佳。Objective To observe the effects and pharmacological effects cf Jiaweixiaoyaosan on learning, memory and immune cells in mice and provide evidence for clinical trials. Methods BALB/c mice were randomized into five groups, namely the normal control group, stress group, Xiaoyaosan group, the low-dose Jiaweixianyaosan group and the high-dose Jiaweixiaoyaosan group. Morris water maze was used to study the changes in learning and memory. The levels of CD4 and CD8 were determined using flow cytometry. Results Compared with the stress group, mice on high-or low-dose Jiaweixiaoyaosan and those on Xiaoyaosan showed reduction in mean escape latency (P〈0.01), greater percentage swimming time in the platform quadrant (P〈0.01). These groups also showed more CD4 T cells (P〈 0.05), less CD8 T cells (P〈0.05) and lowered CD4/CD8 ratio in spleen suspension compared with the stress group (P〈 0.05). Conclusion Xiaoyaosan or Jiaweixiaoyaosan may remarkably ameliorate the impaired learning and memory caused by chronic stress. The beneficial effects were more evident with high-dose Jiaweixiaoyaosan.
