摘 要:目的探讨小肽缀合物(NNP)对心肌缺血/再灌注(I/R)损伤保护作用。方法大鼠左冠状动脉阻塞30min,然后再灌注120min。NNP(10mg/kg)或溶剂在缺血模型形成之前10min经颈静脉给药。再灌注结束后,快速取出心脏进行生化测定和病理组织学分析。结果缺血再灌注损伤之后溶剂组丙二醛(MDA)水平(144±10)nmol/gpro,还原型谷胱甘肽(GSH)水平(460±23)nmol/gpro;NNP组MDA水平(50±30)nmol/gpro,GSH水平(789±137)nmol/gpro;另外,NNP给药组心肌的梗死面积(17±2)cm3,溶剂组(23±3)cm3,2组差异均有统计学意义(P<0.05)。结论NNP可使梗死面积明显减小,并明显改善心脏抗氧化能力,从而缓解脂质过氧化造成的缺血再灌注损伤。Objective To explore the protective effects of nirtonyl nitroxide containing peptide (NNP) on myocardial ischemia reperfusion injury in rats model. Methods Wistar rats were subjected left main coronary artery occlusion for 30 rain, followed by 120 min reperfusion to induce myocardial ischemia reperfusion injury. The NNP (10 mg/kg) or solvents was injected 10 min before ischemia via jugular vein. At the end of reperfusion, the heart was dissected quickly and biochemistry indexes were detected, the pathological histology results were analyzed. Results It showed that the NNP significantly reduced the level of malondialdehyde (MDA, 50±30 nmol/g pro) and increased the level of reduced glutathione (GSH,789 ± 137 nmol/g pro),compared to sham group (MDA, 144±10 nmol/g pro ; GSH , 460 ± 23 nmol/g pro).In addi tion,after the NNP administration,the infarct size ( 17±2)cm^3 significantly reduced than sham group(23 ±3)cm^3 ,there were significant differences betweet the two groups ( P 〈 0.05). Conclusion The results indicated that the NNP could improve the antioxidant capacity of heart and attenuates the degree of lipid peroxidation after myocardial ischemia reperfusion.
