摘  要:目的建立小鼠胚胎干(ES)细胞株S8在无饲养层的条件下短期内连续传代体系,获得大量纯的ES细胞。方法ES-S8细胞无饲养层连续传代后,检测其未分化指标以及多分化潜能。结果ES-S8细胞在无饲养层条件下培养5代,AKP染色、SSEA-1免疫荧光、OCT-4均显阳性,以及经拟胚体(EB)途径自发分化后,能形成多种类型的细胞。结论ES-S8细胞能够在无饲养层条件下培养,至少传5代仍能保持未分化特异性指标和多分化潜能。Objective To establish the method of short-term cultures of mouse embryonic stem cells S8 without feeder layer cells and obtain robust pure ES-S8 cells. Methods ES-S8 cells were cultured for five passages without feeders. The undifferentiated state was confirmed by expression of alkaline phosphatase,SSEA-1 and Oct-4, and the pluripotency of EBs analysed. Results ES-S8 cells were positive for AKP, SSEA-1 and Oct-4 without feeders, and differentiated into a variety of cell types. Conclusion ES-S8 can grow normally at least 5 passages without feeders, and maintain their undifferentiated characteristics and pluripotency.
