摘 要:对小鼠朊蛋白(prp105-125缺失)的基因片段进行扩增,并将扩增片段导入诱饵载体pSos中构建酵母双杂交诱饵载体pSos-prp朊蛋白(prp105-125缺失),用重组质粒转化感受态酵母菌cdc25H,检验其表达产物在酵母细胞中有无毒性及自激活作用。序列分析结果表明,该试验成功构建了小鼠朊蛋白(prp105-125缺失)酵母双杂交诱饵载体,且该诱饵质粒的表达产物对酵母菌cdc25H既无毒性也无自激活作用。The gene segment of prion (prp105-125 flaw) of mice was amplified, and it was transferred into bait vector pSos to constructed bait vector pses-prp with prion (prp105-125 flaw)of two-hybrid system of yeast. The toxicity and self-activation ability of the expression product of recombinant plasmid in yeast cell were detected when it was transferred into yeast cdc25H. The sequence analysis results showed that the bait vector pSos-prp with prion (prp105-125 flaw)of two-hybrid system of yeast was successfully established and its expression product had no toxicity and self-activation ability on yeast cdc25H.
