摘  要:目的:观察鞘内泵入布托啡诺对慢性压迫性损伤(CCI)大鼠脊髓一氧化氮(NO)产量和一氧化氮合酶(NOS),乙酰胆碱酯酶(AchE)活性的影响。方法:清洁级雄性成年SD大鼠32只,随机分为4组,每组术后第4天开始用药,每日1次,连续7d。B组不实施任何手术,但实施同种基础麻醉;C组CCI及鞘内置管后鞘内泵入生理盐水10μL/d;TS组CCI及鞘内置管后鞘内泵入布托啡诺12μg/d;TB组CCI及鞘内置管后鞘内泵入布托啡诺24μg/d。1周后断头处死测NO产量、NOS和AchE活性。结果:C、TS、TB3组脊髓NO产量、NOS活性均明显高于B组(P<0.05),但TS、TB两组脊髓NO产量、NOS活性均明显低于C组(P<0.05),TS、TB两组间脊髓NO产量、NOS活性差异无显著性(P>0.05);B、TS、TB3组脊髓AchE活性均明显低于C组(P<0.05),B、TS,TB3组间脊髓AchE活性差异无显著性(P>0.05)。结论:鞘内泵入布托啡诺能够抑制CCI大鼠脊髓NO产量和NOS、AchE活性升高,是其脊髓水平抗伤害性作用的机制之一。Objective To explore the effects of intrathecal butorphanol on NO production and activities of NOS and AchE in the spinal cord in rats with chronic constriction injury (CCI). Methods 32 healthy male SD rats were randomly divided into 4 groups. Group B received the same basic anesthesia but no surgery. The routine model of CCI was established in the remaining 3 groups. 40 days after the procedure, Group C received 10μL/d of intrathecal normal saline daily for 7 days, group TS received 12 μg/d of butorphanol, and group TB group received 24 μL/d of butorphanol. NO production and activities of NOS and Ache in the spinal cord were measured one week later. Results NO production and NOS activity were significant higher in groups C, TS, and TB than in group B (P 〈 0.05) and were lower in groups TS and TB than in group C (P 〈 0.05) ; they did not differ between group TS and group TB (P 〉 0.05). AchE activity was markedly lower in groups B, TS, and TB than in group C (P 〈 0.05) and did not differ among the first three groups (P 〉 0.05). Conclusion Intrathecal butorphanol can inhibit the increase in both NO production and activities of NOS and AchE, which is one of the mechanisms for the protection against spinal injury.
