摘  要:目的:研究3类新疆地产植物金属硫蛋白(MT)对重金属中毒后机体脂质过氧化的影响,并分析各类植物MT之间有无交互作用,为开发新型重金属中毒拮抗剂——金属硫蛋白提供理论依据。方法:选取经筛选后MT含量较高的植物,按不同种属分为3类,不同种属植物MT作为3个研究因素,各类植物MT原提取液为第一水平,提取液空白溶剂作为第二水平,采用三因素两水平正交实验设计,选用L8(23)正交表,并按该表因素处理原则对急性镉中毒小鼠进行灌胃干预,以血液超氧化物歧化酶(SOD)活力、谷胱甘肽过氧化物酶(GSH-Px)活力和丙二醛(MDA)含量为分析指标,分析三类植物MT的主效应和各级交互作用。结果:(1)3类植物MT提取液主效应对血液SOD活力、MDA含量的影响差异无统计学意义(P>0.05);(2)第一类和第二类植物MT交互作用对血液SOD活力、MDA含量的影响差异有统计学意义(P<0.05);(3)第三类植物MT提取液主效应对血液GSH-Px活力的影响有统计学意义(P<0.05),第二类和第三类植物MT交互作用对血液GSH-Px活力的影响差异有统计学意义(P<0.05)。结论:第一类和第二类配方植物MT对SOD活力的增强和MDA含量的降低存在明显交互作用;第三类植物MT对GSH-Px活力的增强作用优于其他两类,第二类和第三类植物MT对GSH-Px活力增强存在明显交互作用。Objective. Study the interaction of the metallothionein (MT) in three categories of Xinjiang local plants extract on heavy metal poisoning animal model in terms of lipid per oxidation, provide a theoretical basis for exploring a new type of heavy metal poisoning antagonist-metallothionein. Methods. The plants with higher content of MT were screened out and divided into three categories which acted as three factors, MT extract in these three categories of plants was defined as the first level and the blank solvent as the second level, then we employed orthogonal design of three factors and two levels and selected L8 (2^3) orthogonal form. After the acute cadmium poisoning mice were treated with intragastric administration according to the principle of the orthogonal form, we adopted the SOD vitality, GSH-Px and MDA content in blood, liver and kidney as the analysis target, using the orthogonal data analysis module of SPSS 12.0 soft-ware for data processing and finally analyzed the main effect of MT in three categories of plants and their interaction at all levels. Results: (1) The main effects of MT extractive in three categories of the plants on liver SOD vitality, MDA content and GSH-Px activity were not statistically significant (P 〉0.05) ; (2) The interaction between category Ⅰ and Ⅱ plants on liverMDA content was statistically significance (P 〈 0.05);(3) The main effect of MT extract in category Ⅲ plant on the blood GSH-Px activity was statistically significant (P 〈0.05), the interaction between category Ⅱ and Ⅲ plants on blood GSH-Px activity was statistically significance (P 〈0.05). Conclusions: The category I and Ⅱ plant significantly interact in enhancing the vitality of SOD and reducing the content of MDA. The third category of the plant can better enhance the vitality of GSH-Px than the other two categories and there is an obvious interaction between category Ⅱ and category m plants in enhancing the vitality of GSH-Px.
