人体腹部脂肪组织解偶联蛋白1基因表达水平的探索研究

摘  要:目的:哺乳动物能量的消耗主要通过棕色脂肪组织的产热功能实现,其发生机制源于一种特异的蛋白,解偶联蛋白1(uncoupling protein1,UCP1)。而UCP1仅在棕色脂肪细胞中的线粒体内表达,可作为棕色脂肪的标志物。本研究即通过探索成人腹部脂肪(包括皮下和内脏)组织中是否存在有UCP1,进一步探讨成人腹部脂肪组织中UCP1的基因表达水平与成人脂代谢、体质指标、基础代谢的相关关系。方法:研究样本取自2008年8月~2009年1月期间在新疆维吾尔自治区人民医院普外二科需进行开腹外科手术的住院病人,应用RT-PCR技术检测人体腹部脂肪组织中UCP1的基因是否存在表达及表达水平。进一步分析成人腹部皮下、内脏脂肪组织中UCP1 mRNA基因表达量与身高、体重、体质指数、腰臀围、收缩压、舒张压、空腹血浆血糖、总胆固醇、甘油三酯、高密度脂蛋白、胰岛素抵抗指数及基础代谢的相关关系。结果:25例研究对象(男性13例)皮下脂肪组织中与内脏脂肪组织中UCP1/β-actin灰度值比值差异无统计学意义(P=0.822)。相关性分析仅发现有胰岛素抵抗指数(HOMA-IR)与UCP1 mRNA基因表达量有弱相关关系(r=-0.700,P=0.036)。结论:本研究证明了成人腹部脂肪中确实有UCP1的基因表达;此次研究没有发现UCP1基因表达量与BMI、腰围、血压、血脂有关,仅发现与胰岛素抵抗指数有弱相关。腹部脂肪堆积能够引起的胰岛素抵抗,本研究结果提示其机制可能与腹部脂肪组织中棕色脂肪的存在及其数量有关。该结论仍需进一步研究证实。Objective.. Brown adipose tissue (BAT) thermogenesis in mammals contributes to energy expenditure. Uncoupling protein 1 (UCP1) is exclusively expressed in brown adipocytes of mitochondria. The current study aims to investigate if BAT exist or not in white adipose tissue (WAT), by examining the existence of UCP1 in the abdominal fat (including subcutaneous and visceral fat tissue) of adult human. Additionally, the correlation between the gene expression level of the UCP1 with the levels of the adult lipid profiles, body mass index (BMI), basic metabolism, waist circumference and the gene expression level of the UCP1 in the abdominal fat of adult were analyzed. Methods: Twenty-five inpatients were re2 cruited from the People's Hospital of Xinjiang Uygur Autonomous Region who was going to receive abdominal surgeries during the August 2008 to January 2009. Three grams fresh adipose tissue was obtained with informed consent during their surgeries from subcutaneous and visceral fat tissue respectively. We used RT-PCR technology to inspect the existence of expression and expressive quantity of gene of UCP1 in the abdominal WAT of adult. Then the correlation between the level of UCP1 expression in the abdominal WAT of adult and BMI, height, weight, waistline, blood pressure, total cholesterol, triglyceride, fasting glucose, high density lipoprotein cholesterol, and insulin resistance index (HOMA-IR) were analyzed. Results: In total 25 subjects (13 males) with the mean age 44.68±10.93 years (25-65). The gene expression of UCP1, mRNA of UCP1 was successfully amplified in 21 samples. There was no statistic difference (P =0. 822) in the rate of gray scan value between the UCP1/β-actin between subcutaneous and visceral WAT. However, only a weak correlation was identified between HOMA-IR and level of UCP1 expression (r =-0. 700,P =0. 036). Conclusions.
