摘  要:目的:建立以高效液相色谱串联质谱电喷雾检测(LC-MS/MS)法测定人血浆中西替利嗪浓度的方法。方法:以甲醇沉淀蛋白处理血样后进样分析。其中,色谱柱为AllureC18,流动相为2mmol·L-1醋酸胺+0.5%甲酸水溶液∶甲醇=65∶35,流速为0.3mL·min-1,内标为格列齐特;采用电喷雾离子化源,检测方式为正离子多离子反应监测,用于定量分析的离子反应分别为m/z389.2→m/z201.3(西替利嗪)、m/z324.1→m/z127.2(格列齐特)。结果:西替利嗪血药浓度在2.0~1000.0ng·mL-1范围内线性关系良好(r=0.9995);方法回收率≥97.2%,日内RSD≤2.4%,日间RSD≤11.1%,平均提取回收率>96.7%。结论:本方法简便、灵敏、准确可靠,可用于测定人血浆中西替利嗪的浓度及其药动学研究。OBJECTIVE: To establish an LC- MS/ MS method for the determination of cetirizine in human plasma. METHODS: The plasma samples were precipitated with methanol for analysis. Allure C18 column was used with the mobile phase consisted of 2 mmol·L^-1 acetic amine plus 0.5% formic acid-methanol (65:35) at a flow rate of 0.3 mL· min^-1 with Gliclazide used as internal standard. Electrospray ionization (ESI) source was applied and operated in positive iron multiple- reaction monitoring mode (MRM). The ionic reactions used for quantitative analysis were m/z 389.2→m/z 201.3 (cetirizine) and m/z 324.1→m/z 127.2 (gliclazide). RESULTS: The linear range of cetirizine was 2.0--1 000.0 ng·mL^-1( r = 0.999 5); the average methodological recovery was greater than 97.2% and the average extraction recovery was greater than 96.7%; the intra- day RSD was no more than 2.4% vs. inter- day RSD was no more than 11.1%. CONCLUSION: The method was proved to be simple, sensitive, accurate and reliable, and it can be used for the determination and pharmacological study of cetirizine in human plasma.
