摘  要:目的:观察环氧合酶-2(cyclooxygenase-2,COX-2)在大鼠偏侧咀嚼牙槽骨及牙周组织中的表达和分布特点,探讨其在牙槽骨吸收、新生和重建中的作用机制。方法:建立大鼠偏侧咀嚼模型,制备不同时期大鼠咀嚼侧牙槽骨标本,行兔抗大鼠COX-2多克隆抗体(1:200)免疫组织化学染色。结果:在正常组牙周膜周围固有牙槽骨中散在血管内皮细胞胞浆呈弱阳性着色;3d实验组咀嚼侧见部分牙槽骨成骨细胞及散在牙周膜细胞胞浆呈较强阳性着色;1w实验组咀嚼侧为阴性着色;2w和4w实验组咀嚼侧见部分牙周膜细胞胞浆局部呈强阳性着色。结论:偏侧咀嚼改变了牙槽骨及牙周组织所处的生物力学环境,进而诱导了体内牙槽骨成骨细胞和牙周膜细胞COX-2的表达。Objective:To determine the expressions and distributions of cyclooxygenase-2 (COX-2),and the possible functions in the alveolar absorptions and remodeling of the rat in unilateral mastication. Method:The model of rat unilateral mastication was established by extracting one side of premolars and molars. The rat alveolar samples at different time periods were collected and the sections were prepared and stained immunohistochemically with polyclonal rabbit anti-COX-2 at a concentration of 1:200. Result:The positive staining was found in the cytoplasm of a few vascular endothelial cells in control, partial alveolar cells and periodontal cells in masticatory sides of experimental rats three days or two weeks after operation, and partial periodontal cells in those of experimental rats after four weeks. The positive staining located in the masticatory sides of experimental groups,while negative staining was found in the those after one week opeartion. Conclusion:The changed mechanical environments of alveolar and periodontium with unilateral mastication would affect COX-2 expressions of rat alveolar osteoblasts and periodontal cells.
