摘  要:为了揭示长白山区中华蜜蜂种型的分子特点,选取其线粒体DNA(mtDNA)非编码区至COII基因段的序列为研究对象,设计并合成了一种理想的用于扩增中华蜜蜂mtDNA非编码区至COII基因段的引物对E2’/H2’,结果可以获得418bp有效的核酸序列;该序列上A+T含量占84.45%,而G+C的含量只有15.55%,序列里长白山区中华蜜蜂与意大利蜜蜂在核苷酸水平上的种间序列相似性为81.71%,并且序列具有丰富的限制性核酸内切酶的酶切位点,其中,52位上的MseI的酶切位点属于长白山区中华蜜蜂的特异酶切位点,53位处出现一个SNP位点,属于由碱基C转换为碱基T的转换型突变,这也进一步证明了蜜蜂mtDNA非编码区是进化速度较快的区域。长白山区中华蜜蜂mtDNA非编码区至COII基因段序列结果已经获得美国国家生物信息(NCBI)网站GenBank的登录号,登录号为FJ494922。To reveal the molecular traits of Chinese bee (Apis cerana cerana) in Changbai Mountain areas, mtDNA fragment from Non-coding region to COII gene region was amplified, sequenced and analyzed. The results: A pair of primers, E2' and H2', were designed and synthesized. They were better primers to amplify the mtDNA fragment. The sequence amplified with E2'and H2' including 418bp. A+T content was 84.45%, G+C content was only 15.55 %. The similarity of sequences between Chinese bee and western bee (Apis mellifera ligustica) was 81.71%. Abundant restriction endonuclease sites were found in the sequence. A unique site of MseI enzyme appears at locus 52 of the sequence, so that a SNP site appeared at locus 53. The mutation type was C→T base conversion. This also revealed that Non-coding region evolved rapidly. This sequence had been submitted to GenBank of NCBI and its access number was FJ494922.
