摘  要:目的研究银杏苦内酯B(BN52021)对脂多糖(LPS)诱发的急性肺损伤小鼠肺组织中核转录因子κB(NF-κB)蛋白质表达的影响。方法健康昆明小鼠50只,随机分为2组:LPS组和BN52021预处理组。每组又根据不同的观察时间点(0、1、3、9 h和12 h)随机分为5个亚组,每组5只小鼠。通过检测肺湿质量/干质量值(W/D)初步判断肺功能,普通光镜观察HE染色肺组织的病理变化,Western blot检测NF-κB p65在蛋白水平上的表达差异,ELISA法测定肺组织匀浆中肿瘤坏死因子-α(TNF-α)及白细胞介素-10(IL-10)浓度。结果LPS注射后,LPS组W/D显著升高,提示存在肺水肿;BN52021预处理组在不同时间点的W/D均低于LPS组(P<0.05,P<0.01)。LPS注射后1 h即有明显炎症反应的病理变化:两组光镜下均显示肺泡出血及中性粒细胞浸润,但BN52021预处理组明显轻于LPS组。LPS组NF-κB p65呈双峰表达,峰值分别为LPS注射后1 h和9 h;BN52021预处理组NF-κB p65呈单峰表达,峰值出现于9 h,除9 h外,各时间点BN52021预处理组NF-κB p65的表达均低于LPS组(P<0.05)。与LPS组相比,BN52021预处理组可以明显降低TNF-α的表达(P<0.05),但2组IL-10峰值无统计学意义。结论BN52021通过抑制NF-κB的早期活化抑制炎症反应,减轻肺组织损伤。Objective To investigate the effect of ginkgolide B ( BN52021 ) on the NF-kB expression in pulmonary tissue of mice with LPS-induced acute lung injury. Methods Fifty Kunming mice were divided into 2 groups at random: LPS group (L group) and BN52021 pretreated group (B group), and then each group was subsequently divided into 5 subgroups according to different time point (0, 1, 3, 9 and 12 h). The mice from corresponding groups received an intraperitoneal injection of normal saline or 20 mg/kg BN52021 followed by an injection of 15 mg/kg LPS 5 min later. The mice were sacrificed in 0, 1, 3, 9 and 12 h after LPS injection. The wet/dry ratio (W/D) was recorded for pulmonary function and the pathological changes of lung tissues stained by HE were examined by light microscopy. The expression of NF-kB p65 was detected by Western blotting, and the levels of tumor necrosis factor-α (TNF-α) and interleukin-10 (IL-10) were determined by ELISA. Results The significant increase of W/D was found after LPS injection in L group, which indicated the existence of pulmonary edema; while the W/D in B group was lower (P 〈0. 05 or P 〈0. 01 ). The pathological changes of inflammation, such as puhnonary hemorrhage and the inflammatory cells infiltration, were obvious in 2 groups, but that was better in B group. Moreover, the expression of NF-kB p65 displayed double peak at 1 h and 9 h in L group, while there was only one at 9 h in B group. What' s more, the value in B group decreased in comparison with L group (P 〈 0. 05 ) except for the value at 9 h. At last, the level of TNF-α in B group was lower than in L group, but there was no difference of IL-10 between 2 groups. Conclusion BN52021 can induce the inflammation of acute lung injury induced by LPS via inhibiting the early activation of NF-kB.
