摘  要:目的:通过观察青光安颗粒含药血清对加压后人小梁细胞Caspase-3和Caspase-8表达的影响,探讨青光安颗粒含药血清抑制加压后人小梁细胞凋亡的分子机制。方法:制备青光安颗粒2.5,5,10,20倍含药血清、益脉康5倍含药血清和不含药血清,建立人小梁细胞体外加压模型,用不同药物与浓度的含药血清分别作用体外加压培养人小梁细胞,用TUNEL技术及流式细胞术检测各组的凋亡细胞。用免疫组化、流式细胞术分析加压后人小梁细胞Caspase-3和Caspase-8表达的变化。结果:青光安颗粒5,10,20倍组和益脉康组都可以显著抑制加压后人小梁细胞的凋亡和Caspase-3,Caspase-8的表达,与空白组比较,差异有显著统计学意义(P<0.01);青光安5倍组与益脉康5倍组比较差异无统计学意义(P>0.05);与青光安10倍组比较,青光安5倍组、益脉康5倍组的细胞凋亡率和Caspase-3和Caspase-8表达率都较高,比较差异都有统计学意义(P<0.05),但青光安10倍组与青光安20倍组比较,差异无统计学意义(P>0.05)。结论:青光安颗粒通过阻断Caspase-3与Caspase-8凋亡信号传导系统而抑制加压后人小梁细胞的凋亡。AIM: To observe the effects of serum containing qingguang'an grains on the expression of Caspase-3, Caspase-8 of pressurized human trabecular cells cultured in vitro, And to investigate the molecule mechanism of serum containing qingguang'an grains in inhibiting apoptosis of pressurized human trabecular cells. METHODS: Serums containing 2.5, 5, 10 times and 20 times qingguang'an grains, serum contain-ing 5 times yimaikang tablets and serum-free were prepared; the model of pressurized human trabecular cells cultured in vitro was installed; the pressurized human trabecular cells were incubated with serum contain-ing different medicines and concentrations; the apoptotic cells were determined by TUNEL technique and flow cytometry;caspase-3 and Caspase-8 proteins were measured by immunohistochemistry and flow cytometry. RESULTS: 5, 10 times and 20 times qingguang'an grains groups and yimaikang tablets group could inhibit the apoptosis and the expression of Caspase-3, Caspase-8 of pressurized human trabecular cells obviously. As compared with the blank group , the apoptosis and the expressions of Caspase-3 and Caspase-8 were lower( P 〈 0.01 ) ; there were no obvious difference between 5 times qingguang'an grains group and 5 times yimaikang tablets group( P〉 0.05); as compared with the 10 times qingguang'an grains group ,the apoptosis and the expressions of Caspase-3 and Caspase-8 of 5 times qingguang'an grains and 5 times yimaikang tablets groups were higher (P 〈 0.05), But there were no obvious difference between 10 times qingguang'an grains group and 20 times qinggu- ang'an grains group(P〉0.05). CONCLUSION : Caspase-3 and Caspase-8 play important roles in apoptosis of pressurized human trabecular cells, qingguang'an grains can effectively inhibit apoptosis of pressurized human trabecular cells by influenced the signal transduction system of Caspase-3 and Caspase-8 probably.
