摘 要:目的观察细胞凋亡在急性肺损伤(ALI)幼猪肠黏膜上皮的发生情况及相关机制。方法采用脂多糖(LPS)诱导幼猪ALI模型,分离肠黏膜上皮细胞,采用流式细胞结合末端脱氧核苷酸转移酶介导的dUTP原位缺口末端标记(TUNEL)和电镜方法观察凋亡细胞的改变。同时检测炎症/抗炎因子TNF-α/IL-10 mRNA的表达和氧化与抗氧化损伤指标丙二醛(MDA)、超氧化物歧化酶(SOD)水平与凋亡发生的关系,进一步阐明凋亡的发生机制。结果模型组肠上皮细胞凋亡较正常组明显增加(P<0.01);模型组TNF-α mRNA表达增强,IL-10 mRNA表达减弱,MDA含量增加、SOD活性减低与正常组比较差异均有统计学意义(P<0.01)。结论ALI幼猪肠黏膜上皮细胞凋亡增加,与炎症/抗炎因子表达失衡,氧化/抗氧化损伤异常有关。Objective To investigate the changes and mechanisms of apoptosis of the intestinal mucosa epithelial cells in piglet with acute lung injury (ALI). Methods The model of ALI piglet was induced by lipopolysaccharide (LPS). Intestinal mucosa epithelial cells were isolated and tested with flow cytometry, TUNEL and electron microscope were used to detect the changes of apoptosis. The expression of TNF-cdIL-10 mRNA (inflammatory and anti-inflammatory factors), marker of oxidation and anti-oxidation, MDA, and SOD, respectively, in the intestinal tissue were measured. Results In comparison with the normal group, the number of apoptotic cells increased significantly, TNF-α mRNA expression and the level of MDA were significantly higher, while IL-10 mRNA expression and SOD activity were obviously lower in the model group (all were P 〈 0.01 ). Conclusions The apoptotic intestinal enterocytes increased in the piglet with ALI, related to the imbalance of inflammatory/anti-inflammatory factors and the pathological changes of oxidation/ anti-oxidation.
