还脑益聪方对老龄认知障碍大鼠脂质代谢和海马内质网Aβ相关结合蛋白表达的影响  

Effect of Huannaoyicong Prescription to AD Rat in Learning and Memory and Hippocampus of Aβ-related Protein Expression with Lipid Metabolism

摘　　要：目的:观察还脑益聪方对老龄认知障碍-痴呆(AD)大鼠学习记忆、脂质代谢和海马内质网Aβ相关结合蛋白(ERAβ)表达的影响。方法:利用Morris水迷宫筛选老龄认知障碍大鼠40只,按随机表分为还脑益聪方大剂量(13.68g生药/kg)组、小剂量(6.84g生药/kg)组、阳性药物对照组(盐酸多奈哌齐片,0.45mg/kg)组和模型(认知障碍老龄大鼠)对照组,每组10只,灌胃给药8周,观察大鼠学习记忆行为,血脂指标和海马组织病理形态及免疫组化ERAβ表达的变化。另设健康青年组大鼠10只和健康老年组大鼠10只作为对照组。结果:与青年组大鼠比较,老龄认知障碍大鼠的血脂水平增高(P<0.05),海马组织病理形态改变明显,ERAβ表达显著增高(P<0.01)。与老龄认知障碍模型大鼠比较,还脑益聪方能显著缩短AD大鼠寻台时间和寻台路线(P<0.01,P<0.05),降低血清胆固醇的水平(P<0.01),改善大鼠海马组织的病理形态,抑制大鼠海马ERAβ的表达。结论:还脑益聪方具有调节老龄痴呆大鼠的血脂水平,抑制ERAβ的表达,从而减轻海马区神经元损伤,提高老龄认知障碍大鼠的学习记忆能力。Objictive：To approach effect of Huannaoyicong prescription（HNYC） to AD rat in ethology and Endoplasmic reticulum Aβ-related protein （ERAβ） expression, hippocamp tissue pathomorphology and Lipid metabolism. Methods：The AD aged rat model was applied, and the rats were divided to high and low HNYC prescription treating group, Donepezil Hydrochloride Tablets control group, aging rat model group, young rat blank group and aging rat blank group. Index change of ethology, hippocamp tissue pathomorphology and ERAβ expression with Immunohistochemical method, Lipid Determination were tested. Results： Time and distance of AD rats look for platform were shorten obviously, and Increase hippocamp tissue ERAβ expression, hippocamp nerve cell injury and Lipid targets change were improved by high and/or low HNYC prescription. And it was superior to those of aging rat model group（ P 〈 0.01, P 〈 0.05）. Conclusion： The results showed that the of Huannaoyicong prescription is enhance learning and memory ability in AD rats. Partly mechanism is that it can improve Lipid Determination and Inhibition ERAβ expression of Brain tissue. There are pharmacological effects with HNYC prescription in AD.
