摘 要:目的:选取HIF-1α基因第12外显子上的C1772T单核苷酸多态(SNP/C1772T)解析,并与有氧耐力训练后左心室结构、功能指标进行关联性研究,探讨该多态位点作为预测有氧耐力训练后左心室结构、功能指标适应性改变的分子遗传学标记的可行性。方法:实验对象为102名中国北方士兵。受试者进行18周有氧耐力训练,测试有氧耐力训练前后安静及递增负荷50 W、100 W、150 W下左心室结构、功能等指标,应用PCR-RFLP解析HIF-1α基因SNP/C1772T的基因型,采用关联(Association-Study)研究方法,研究18周有氧耐力训练前后安静及递增负荷左心室结构、功能等指标变化量与HIF-1α基因SNP/C1772T关联性。结果:1)HIF-1α基因SNP/C1772T基因型在士兵中分布频率经卡方检验[x^2(df=2) =1.05,P>0.05]符合Hardy-Weinberg平衡;2)SNP/C1772T的CC基因型受试者18周有氧耐力训练后安静状态下左心室结构指标,左心室重量指数(left ventricular mass index,LV-MI)、左室舒张末期内径(left Ventricular diastolic internal dimension,EDD)、舒张未期左室后壁厚度(left ventricular posterior wall diastolic,PWD)等变化量与CT基因型受试者相比表现出明显改善(P<0.05,P<0.10),其增长率分别为9%,4%,2%;左心室功能指标,每搏输出量(stroke volume,SV)、每搏指数(stroke volume index,SVI)、射血分数(ejection fraction,EF)、心动周期(cardiac cycle,T)变化量也表现为高于CT基因型的趋势(P<0.1);3)SNP/C1772T的CC基因型受试者18周有氧耐力训练后,递增负荷实验150 W时CC基因型受试者SVI、心功能指数(cardiac output index,COI)的变化量显著高于CT基因型(P<0.05),SV、每分输出量(cardiac output,CO)的变化量有高于CT型的趋势(P<0.10)。结论:HIF-1α基因SNP/C1772T多态性CC基因型可作为预测有氧耐力训练后左心室结构指标LVMI、EDD、PWD和功能指标COI、SVI等敏感性变化的分子遗传学标记。Objective: In present study, we researched the association between single nucleotide polymorphism (SNP) C1772T, which located in exon 12 of HIF-1 Q gene, and left ventricular structure and function, and explored feasibility that SNP/C1772T was as genetic mark for predicting the effect of aerobic endurance training. Method: The 102 soldiers selected in present study who performed aerobic training program 3 times per week for 18 weeks at 95 % individ- ual ventilation threshold intensity. Some phenotype parameters, such as left ventricular structure and function, were measured at the state of resting and gradual increased loads exercising (50 W, 100W and 150 W) before and after aerobic training. Meantime,Genotyping of SNP/ C1772T of all subjects was performed by polymerase chain reaction amplification. Result:De- partures from Hardy-weinberg equilibrium were not statistically significant in SNP C1772T. After 18 weeks aerobic training, left ventricular mass index (LVMI), left ventricular diastolic internal dimension (EDD), left ventricular posterior wall diastolic (PWD) at resting increased separately by 9 %, 4 %, 2 % in subjects with CC genotype, and the changes of these in CC genotype were significant higher than of CT genotype(P〈0.05,P〈0. 10). And the changes of the parameters of left ventricular function such as stroke volume (SV), (stroke volume index (SVI) ,ejection fraction (EF), cardiac cycle (T) in CC genotype indicated higher tendency than those in CT genotype (P 〈 0. 10) in resting. During gradual increased loads experiment, the changes of SV, SVI, CO and COI in CC genotype were significant higher than those in CT genotype in 150W after 18 weeks aerobic training(P〈0.05, P〈0. 10). Conclusion: The re-suit in the present study suggested that SNP/C1772T in HIF-lα gene was a genetic factor that affected the responses in cardiac structure and function to aerobic training.
