摘  要:目的:建立巨噬细胞甘露糖受体(MMR)模型,用于筛选中药中以甘露糖受体(MR)为靶标的活性物质。方法:将小鼠腹腔巨噬细胞分别与不同浓度的D-甘露糖和D-半乳糖共孵,用流式细胞仪和荧光显微镜检测D-甘露糖和D-半乳糖对Mφ结合异硫氰酸荧光素标记的甘露糖化牛血清白蛋白(M-FITC-BSA)的拮抗作用,优化实验条件,建立MMR筛选模型;将上述模型用于筛选四物汤等6个复方多糖中的MR结合成分。结果:两种检测方法均显示:随D-甘露糖浓度的上升,M-FITC-BSA标记的Mφ检出率逐步下降(P<0.01),而这种现象在D-半乳糖组中未观察到;当D-甘露糖达到0.080mmol/L浓度时,便能明显拮抗Mφ结合M-FITC-BSA(P<0.05或P<0.01)。复方多糖的筛选结果显示:在一定浓度下,四君子汤、六味地黄汤和玉屏风散多糖组分能明显拮抗小鼠腹腔Mφ与M-FITC-BSA的结合,且具有浓度组间差异性(P<0.05或P<0.01)。结论:MMR模型对MR结合成分的筛选作用稳定而敏感,用于中药MR结合物的筛选,对快速筛选中药免疫调节成分、研究中药免疫调节机制具有重要意义。AIM : To establish macrophage mannose receiver Model (MMR), and use it to screen active cornponent with mannose receiver (MR) as target from traditional Chinese herbs. METHODS: The mouse abdominal macrophages was hatched with D-mannose and D-galactose of the different concentration, and the flow cytometry and fluorescence microscope were used to measure the antagonistic effect of M-FITC-BSA ( Mannose-fluorescein iso- thiocyanate-bovine serum albumin) with D-mannose and D-galactose. After the MMR screening model was estab- lished to screen MR union ingredients of polysaccharide ingredients from six compound prescriptions, such as Siwu Decoction and so on. RESULTS : Both of measuring methods showed that when D-mannose concentration increased the relevance ratio of Mψ marked with M-FITC-BSA decreases (P 〈 0.01 ). However, this phenomenon did not ap- pear in case of the D-galactose group. When the concentration of D-mannose reached 0.080 mmol/L, it could obvi- ously rivalry Mψ to combine with M-FITC-BSA (P 〈0.05 or P 〈0.01 ). The result of screening of compound pre- scription polysaccharide demonstrated that Sijunzi Decoction, Liuweidihuang Decoction and Yupingfeng Powder polysaccharide, under certain concentration, could obviously inhibit mouse abdominal Mψ to combine with M-FITC-BSA, and they had difference at some concentration groups ( P 〈 0.05 or P 〈 0.01 ). CONCLUSION: Screening function of MMR model for ingredient combined with MR is stable and sensitive. When being used to traditional Chinese herbs, it will play an important part in fast screening of immunity adjustment ingredients and immunity adjustment mechanism.
