摘  要:目的研究小剂量饮酒对四氯化碳(CCl4)诱导的小鼠肝纤维化的影响。方法40只雄性昆明小鼠随机分为4组,空白对照组小鼠自由饮水,予植物油皮下注射;四氯化碳模型组小鼠自由饮水,予以CCl4皮下注射;酒精模型组小鼠予酒精代水饮,并皮下注射植物油;酒精+四氯化碳组小鼠予以酒精代水饮,并皮下注射CCl4。通过检测小鼠血清ALT、AST水平,肝脏的病理组织学改变及肝组织转化生长因子-β1(TGF-β1)mRNA和I型胶原(collagen I)mRNA的表达水平以观察小剂量饮酒对CCl4诱导的小鼠肝纤维化的影响。结果酒精+四氯化碳组小鼠与其他3组小鼠相比,血清ALT、AST水平,肝细胞坏死程度和肝纤维化程度以及TGF-β1、collagen I mRNA的表达水平均为最高。结论小剂量饮酒加重了CCl4诱导的小鼠肝纤维化和肝功能损伤程度。Objective To observe the effect of light alcohol consumption on carbon tetrachloride (CCl4) -induced liver fibrosis in mice. Methods Forty Kunming mice were randomly divided into 4 groups: a normal control group had free access to tap water and were given plant oil through subcutaneous injection; a CCl4 model group had free access to tap water and were given CCl4 through subcutaneous injection; an alcohol model group had free access to 10% alcohol as drinking water and were given plant oil through subcutaneous injection; and an alcohol plus CCl4 group had free access to 10% alcohol as drinking water and were given CCl4 through subcutaneous injection. Mice in each group were sacrificed in 8 weeks. The level of serum ALT, AST, histopathology of the liver and mRNA expression of TGF β1 and collagen I was detected. Results The level of serum ALT, AST, degree of hepatic inflammation and fibrosis, and the mRNA expression of TGF β1 and collagen I of mice in the alcohol plus CCl4 group were all the highest. Conclusion Light alcohol consumption can aggravate CCl4-induced liver fibrosis.
