重组人结缔组织生长因子对肾小管上皮细胞表达α5β1整合素及纤连蛋白的影响

摘　　要：目的:研究在不同情况下,重组人结缔组织生长因子(rhCTGF)对肾小管上皮细胞(HK-2)表达α5β1整合素及纤连蛋白(F ibronectin,FN)水平的影响。方法:rhCTGF以0、50 ng.mL-1干预HK2,采用免疫细胞化学检测,观察α5β1整合素的表达情况。rhCTGF以0、5、50 ng.mL-12天干预HK-2,观察rhCTGF对纤连蛋白的表达影响的浓度效应;rhCTGF以10 ng.mL-1于0、24、48、72小时干预HK-2,观察rhCTGF对纤连蛋白的表达影响的时间效应;免疫印记检测纤连蛋白的表达水平。结果:在基础状态下,肾小管上皮细胞仅有少量α5β1整合素表达,50 ng.mL-1rhCTGF可使α5β1整合素表达量显著增加(P<0.05)。在基础状态下,人近端肾小管上皮细胞表达少量纤连蛋白。5 ng.mL-1rhCTGF可使纤连蛋白表达量显著增加(P<0.05),随着rhCTGF浓度增加,蛋白表达也进一步增加,呈现浓度依赖性变化效应。rhCTGF刺激24 h,近端小管上皮细胞纤连蛋白表达开始增加,48、72 h进一步增加,呈现时间依赖效应。结论:rhCTGF能使近端小管上皮细胞表达α5β1整合素水平显著增加。rhCTGF呈时间及浓度依赖性诱导人近端肾小管上皮细胞纤连蛋白表达。Objective： To observe the effects of rhCTGF on the expression of α5β1 integrin and fibronectin in renal tubular epithenial cells. Methods：Human tubular epithelial ceils （HK-2）were treated with rhCTGF of 0 and 50 ng·mL^-1 to test the expression of α5β1 integrin by immunocytochemistry. HK-2 was treated with different concentration of rhCTGF （0, 5, 50 ng·mL^-1 ） for different time （10 ng·mL^-1 rhCTGF treated for 0, 24, 48, 72 hours） to test fibronectin by Western blot. Results： rhCTGF 50ng·mL^-1 group markedly improved HK-2 expressed α5β1 integrin compared with the control. rhCTGF induced increase of the expression of fibronectin in a dose and time dependent manner. After 48 hours of exposure to the increased rhCTGF concentrations, the fibronectin protein expression was progressively upregulated. After exposure to rhCTGF （10 ng·mL^-1） in 0, 24, 48, 72 hours, the fibronectin protein expression was progressively upregulated compared with control （P 〈 0.05 ）. Gonclusion ： rhCTGF could upregulate the expression of α5β1 integrin and induce the increase of the expression of fibronectin in a dose and time dependent manner in renal proximal tubular epithenial cells.
