摘 要:目的:筛选与精子活力相关的基因。方法:将活力正常者与弱精子症者精子cDNA与Affymetrix全基因组芯片进行杂交,筛选出差异表达基因。RT-PCR分析该基因在人体不同组织及精子中的表达,并比较该基因在活力正常和活力低下的人精子中表达的差异。结果:芯片结果分析筛选出差异表达基因:电压依赖性阴离子通道(VDACs)基因。VDACs包括VDAC1、VDAC2、VDAC33个亚型,它们均在人精子中表达。与活力正常组精子VDAC2相对表达量(0.803±0.043)比较,VDAC2在弱精子症组精子的相对表达量(0.568±0.036)显著降低(P<0.01)。结论:VDAC2的表达减少可能与精子中活力降低有关。Objective: To identify the genes involved in sperm motility. Methods: We hybridized asthenospermial and normal motile sperm cDNA samples with the human whole genome Affymetrix chip to screen differentially expressed genes. Then we detected the mRNA expressions of the voltage-dependent anion channel genes (VDACs) in human organs and spermatozoa by RT-PCR and compared their expressions in the poor and normal motility spermatozoa. Results: Differentially expressed genes VDACs were identified by anal- ysis of the hybridization signals, including the 3 subtypes VDAC1, VDAC2 and VDAC3. The expression of VDAC2 mRNA was significantly decreased in the poor motility sperm (0. 568 + 0. 036), as compared with the healthy men (0. 803 ± 0. 043, P 〈 0.01 ). Conclusion: The decreased expression of VDAC2 in the ejaculated spermatozoa is possibly associated with the reduction of sperm motility.
