摘 要:目的:建立制备免疫刺激复合物(ISCOMs)的方法,ISCOMs是一种疫苗制备新技术,具有突出的免疫学价值。方法:本研究以人巨细胞病毒(HCMV)包膜蛋白和皂苷类佐剂QuilA作用,采用非离子去垢剂Mega-10及超速离心技术,进行了HCMVISCOMs实验型疫苗的制备。结果:浓缩样品液经蔗糖梯度超速离心后,在离心管底部可见白色病毒带;纯化样品电镜观察具有典型的ISCOMs形态。结论:本研究制备样品在电镜下观察见抗原蛋白分布在ISCOMs颗粒表面,为直径30nm左右的球状结构,与国外文献所报道形态相符。Objective: To establish the method for preparing immunostimulating complexes (ISCOMs) which is a new technology for preparing vaccine and has prominent value in immunology. Methods: In this study, the envelope proteins of HCMV were complexed with the glycoside Quil A by hydrophobic interaction. Mega 10 and sucrose gradient centrifugation were also used. Results: Sucrose density gradient centrifugation gave ISCOMs as a white sediment in the centrifuge tube. Electronmicroscopic examination showed that the preparation had the typical appearance and structure of ISCOMs. Conclusion: Under electronmicroscope the ISCOMs were spherical particles of 30 nm in diameter.
