摘  要:目的观察蝎毒多肽提取物(PESV,防治白血病中药提取物)对抑制人白血病原代细胞粘附力的影响。方法以人白血病原代细胞为研究对象,台盼蓝拒染法检测PESV对人白血病原代细胞的细胞毒作用;观察其对白血病原代细胞与人脐静脉内皮细胞(HUVEC)粘附的影响。结果PESV在对细胞最佳作用浓度15μg.mL-1下,能够明显抑制人白血病原代细胞与HU-VEC的粘附,抑制率为(52.18±21.01)%;各浓度组与阳性对照组比较具有显著性差异(P<0.05)。结论PESV在对细胞最佳作用浓度下,对人白血病原代细胞的黏附力有明显抑制作用。Objective To investigate the effect of peptide extract from scorpion venom (PESV) in suppressing adhesion and invasion of leukemic primary cell. Methods By the method of trypan blue dye exclusion, the cytotoxic action on humanistic leukemic primary cell induced by PESV was examined. The effect of PESV on adhesion of leukemic primary cell and human umbilical vein endothelial ceils (HUVEC) was observed. The transwell chamber was used to explore the effect of PESV on invasion of leukemic primary cell. Results The best concentration of PESV, namely 15 μg.mL^-1, could evidently depress the adherent effect of leukemic primary cell and HUVEC, and the inhibition ratio was (52. 18 ± 21.01 )%. Compared with positive control group, other concentration groups could obviously depress the adherent effect ( P 〈 0. 05 ). Compared with positive control group, this concentration of PESV could clearly depress the breakthrough ability against artificial basal lamina of leukemic primary cell ( P 〈 0. 05 ), and the inhibition ratio was (50.07±11.72 ) %. Conclusion The best concentration of PESV has significant inhibitory effect on invasion and transfer of leukemic primary cell.
