Objective:To investigate the effects of the active fraction of Semen Ziziphi Spinosae(SZS) on morphine - induced conditioned place preference (CPP) in mice. Methods : Daily subcutaneous injection of morphine(6 mg·kg^-1 ) was administered to mice for 5 days to induce CPP in mice, and daily treatment with a series of extracts of SZS (ip) was given 30 min prior to the administration of morphine to observe the effect of SZS on morphine -induced CPP. Result: Morphine (6 mg·kg^-1, sc) induced mice to obtain a significant CPP and the CPP model was reliable and stable. By pretreating with alcoholic extract of SZS 800mg·kg^-1 ( but not 200mg·kg^-1 or 400mg·kg^-1 ), ip, before morphine injection, the morphine - induced CPP was antagonized significantly( P 〈 0. 05). Then by pretreating with acetoacetate( 13 mg·kg^-1,20 mg·kg^-1 ), and n - butanol ( 160mg·kg^-1) extract of SZS, the CPP was also inhibited ( P 〈 0. 05 ), but the CPP was not affected by petroleum benzin, ethereal and aqueous extract of SZS. Conclusion : The effects of the active fraction of SZS can suppress the acquisition of morphine induced CPP in mice.
