多巴胺β羟化酶基因-1021C/T多态性与海洛因依赖的相关性分析

摘　　要：目的:探讨多巴胺β羟化酶(DBH)基因-1021C/T多态性与海洛因依赖的相关性。方法:采用病例对照关联分析,应用PCR-RFLP技术检测112例海洛因依赖者和100例正常对照者的DBH基因-1021C/T多态性的基因型分布和等位基因频率并进行关联分析。结果:DBH基因-1021C/T多态性在海洛因依赖组和正常对照组的分布差异无显著性;DBH基因-1021C/T多态性在对照组和海洛因依赖轻型组、中型组及重型组间分布差异无显著性;DBH基因-1021C/T多态性在海洛因烫吸组、静脉注射组分布差异无显著性,但静脉注射组CT/TT基因型比例显著低于烫吸组(P<0.05);在海洛因烫吸者中CC、CT及TT基因型日使用量差异无显著性;在静脉注射者中TT型日用量是CC和CT基因型的日使用量的2倍(P<0.01)。结论:DBH基因-1021C/T多态性与海洛因依赖无明显相关性,但与海洛因依赖者的某些行为特征有关联。Objective： To explore the association between the -1021C/T polymorphism in dopamine β hydroxylase （DBH） gene and heroin dependence. Metho.ds： In this case control study, the DBH gene - 1021C/T allele genotypes of 112 heroin dependent patients and 100 controls were detected by PCR - RFLP, and the association between heroin dependence and polymorphism of DBH gene was analyzed. Results：The frequencies of genotype and alleles at the - 1021C/T polymorphism in DBH gene were not significantly different between the study cases and controls, and the polymorphism showed no significant difference among the mild, moderate, and heavy heroin dependent groups. The polymorphism in the smoking and injection of heroin dependent groups was not significantly different, but the frequencies of genotype CT/TT in the injection group were significantly lower than those in the smoking group （P 〈 0. 05）. The average daily amounts of heroin abuse in the CC, CT and tit genotypes were not significantly different in the smoking group, however, the average daily dose of heroin in the TT genotype was twice as big as that of the other genotypes in the injection group（ P 〈 0. 01 ）. Conclusion：The - 1021C/T polymorphism in DBH gene may have no association with heroin dependence, and has certain association with some behavioral characters of heroin addicts.
