摘 要:目的:了解2008年5~9月广州市卫生行业从业人员中德尔卑沙门菌的耐药情况,并对菌株进行脉冲场凝胶电泳(PFGE)基因分型,探索广州市从业人员中德尔卑沙门菌的分子分型多态性。方法:从健康体检的人群中分离并鉴定为德尔卑沙门菌,对鉴定出的13株菌株,选择18种抗生素用纸片法进行药物敏感试验;裂解菌细胞并释放全基因组DNA,经限制性内切酶Xba I酶切,用脉冲场凝胶电泳进行分子分型,使用Quantity-One TM软件成像并使用BioNumerics4.0软件对其同源性进行分析比较。结果:13株沙门菌对9种抗生素敏感,对其余抗生素出现中敏或耐药。对四环素耐药率最高(76.9%),其次为氯霉素(30.8%),复方新诺明(15.4%),氧氟沙星和氨苄西林(7.7%)。13株受试菌株的DNA分子分型图谱,经软件分析,有两株相似性为100%,其他菌株相似性在73.7%和93.8%之间。结论:德尔卑沙门菌对头孢类抗生素完全敏感,对其他抗生素存在中敏或耐药;菌株之间有较高的同源性。Objective:To survey the status of antimicrobial resistance of Salmonella derby isolated from staff in food - related industry from May to September in 2008 and genotype the above strains by pulsed field gel electrophoresis(PFGE) , exploring the polymorphism of Salmonella derby pulsotypes of these strains. Methods:Thirteen strains of Salmonella derby were isolated and identified from staff in food - related industry subjected to physical examination and antimicrobial susceptibility testing were conducted on these strains with Kirby Bauer methods using 18 kinds of antibiotics. Bacterial cells were lysised to release the whole genomic DNA, digested with restriction endonuclease and then subjected to molecular subtyping utilizing PFGE. Profiles were obtained with Quantity - One TM software and their homological analysis were conducted by BioNumerics 4.0 software. Results: Thirteen strains of Salmonella derby were susceptible to nine antibiotics and exhibited medium or resistance to other antibiotics. These Salmonella derby shown the highest rate of drug resistance to tetracycline(76, 9% ) , followed by ehloroein (30. 8% ) , compound sulfamethoxazole ( 15.4% ), ofloxacin (7.7 % ) and ampicillin ( 7.7 % ). Analysis of the PFGE profiles of the 13 strains of Salmonella derby by BioNumerics 4. 0 software indicated that two of the strains shared the same pulsotype and the similarity coefficient of other strains were between 73.7% and 93. 8%. Conclusion:Salmonella derby is resistant to cephalosporins antibiotics and exhibits medium or resistance to other antibiotics. These strains share relatively high homology.
