摘  要:目的观察甲型流感病毒M2基因与GM-CSF基因DNA疫苗的小鼠免疫效果。方法提取甲型流感病毒DNA疫苗质粒。将BALB/c小鼠随机分成流感裂解疫苗对照组、空载体pVAXI对照组、pVAX/GM-CSF对照组、pVAX/M2实验组、pMIG实验组,按0、2、4程序免疫BALB/c小鼠,第一针免疫后1周,定期对小鼠眼眶采血。用血凝抑制(HI)试验检测特异性抗体,流式细胞术双标法检测脾脏T细胞表面CD4+、CD8+分子数量,ELISA方法检测细胞因子IL-4和IFN-γ的含量,并进行保护性试验。结果流感裂解疫苗对照组HI平均值均>40,产生了特异性血凝素抗体,其余各组均未检测到抗体;流感裂解疫苗组具有较强的刺激淋巴细胞增殖的作用,各组间T淋巴细胞表面CD4+及CD4+/CD8+的水平比较,差异有统计学意义;DNA疫苗组可刺激机体产生较高水平的细胞因子,且保护率较流感裂解疫苗对照组提高了一倍。结论甲型流感病毒DNA疫苗可诱导小鼠的免疫应答,且细胞免疫水平显著高于流感裂解疫苗。Objective To observe the immune effect of DNA vaccine carrying M2 gene of influenza A virus and GM - CSF gene in mice. Methods The plasmid of influenza A virus DNA vaccine was extracted. BALB/c mice were divided into five groups and immunized with influenza virus cleavage vaccine, empty pVAX I , pVAX/GM - CSF, pVAX/M2 and pMIG respectively, on days 0, 2 and 4. Serum samples were collected after immunization and determined for specific antibody by HI test, for counts of CD4 + and CD8 + molecules on surface of splenic T cells by flow cytometry, and for IL - 4 and IFN - -rcontents by ELISA. The protective effect of DNA vaccine was observed. Results Specific HI antibody titer was more than 1 : 40 in influenza virus cleavage vaccine group, but was undetectable in any of the other four groups. Influenza virus cleavage vaccine showed strong stimulating effect on the proliferation of lymphocytes. Significant differences were observed in the CD4^+ T cell counts and CD4^+ / CD8^+ values of various groups. DNA vaccine induced high cytokine level, and its protective rate to immunized mice increased doubly as compared with that of influenza virus cleavage vaccine. Conclusion Influenza A virus DNA vaccine can induce immune response in mice. The cellular immunity induced by the vaccine was significantly higher than that by influenza virus cleavage vaccine.
