摘 要:目的探讨不同浓度内皮素(ET)对培养人心包间皮细胞内钙离子浓度的影响。方法体外培养人心包间皮细胞,以Fluo-3/AM负载细胞,利用激光扫描共焦显微镜测定10-7mol/L、10-8mol/L、10-9mol/L内皮素分别作用于23组间皮细胞后细胞内钙离子浓度的变化。结果心包间皮细胞内钙离子浓度随时间、ET浓度变化出现明显改变(P〈0.001);不同浓度ET与不同时间测定的钙离子浓度之间存在交互作用(P〈0.001)。结论不同浓度内皮素作用于人心包间皮细胞后可引起细胞内钙离子浓度的改变;内皮素可能通过自分泌或旁分泌的方式作用于心包;人心包间皮细胞可能具有重要的生物活性。Objective To investigate the effect of different concentrations of endothelin on intracellular calcium level in cultured human pericardial mesothelial cells. Methods Pericardial mesothelial cells from patients undergoing open heart surgery were cultured in vitro. Cells of passage 8-10 were used in this experiment. When confluence was reached, the mesothelial ceils were loaded with Fluo-3/AM. The intracellular calcium concentrations in theses cultured cells were determined by confocal laser scanning microscopy after 10^-7 mol/L, 10^-8 mol/L, 10^-9 mol/L ET being added respectively. Results Calcium concentrations in cultured mesothelial cells increased remarkably after adding three kinds of ET concentration( P 〈 0.001 ). There was interaction between endothelin concentration and observation time ( P 〈 0.001 ). Conclusion Calcium in cultured mesothelial cells changed distinctly after the stimulation of different concentration of ET; Human pericardial mesothelial ceils may possess important biological activity and endothelin may play an important role in the pericardium in an autocrine and/or paracrine mechanism.
