摘  要:利用农杆菌介导法对低酚棉胚性愈伤组织转化转录因子DREB1C基因,探讨了低酚棉胚性愈伤组织对潮霉素的敏感性和胚性愈伤组织状态对转化效率的影响。结果表明:胚性愈伤组织对潮霉素较为敏感,适宜的筛选浓度为25mg/L,可明显抑制非转化组织生长。以继代培养7~14d的胚性愈伤组织作为转化受体,转化率高达56.3%。将转化后获得的抗性愈伤组织转入分化培养基中分化成苗,经过PCR检测得到5株阳性植株。The DREB1C gene was transferred into embryogenic callus of glandless cotton through Agrobacterium-mediated transformation. The sensitivity of embryogenic callus to hygromycin and the effects of the state of embryogenic callus to transformation efficiency were studied. The results showed that the embryogenic callus was sensitive to hygromycin, and the suitable screening concentration was 25mg/L, which could significantly inhibit the growth of non-transformation. The transformation efficiency reached 56.3% when the embryogenic calli were subcultured for two weeks. The resistant calli were cultured on differentiation medium for embryo maturation and plant regeneration. Five transgenic plants were verified by PCR.
