超声联合微泡与高温热疗开放大鼠血脑屏障的对比研究

摘  要:目的探讨1MHz低频超声诱导微泡破坏和高温热疗两种方法对大鼠血脑屏障通透性的影响。方法将大鼠分为2个实验组与1个对照组,实验A组大鼠经股静脉注入微泡造影剂后,采用频率1MHz、声强1.2W/cm的超声波经大鼠颅骨照射3min;实验B组大鼠于加温仓以36℃加温3h;对照组(C组)仅室温下静脉注射生理盐水。干湿重法测定脑水含量,伊文思蓝测定法观察大鼠血脑屏障通透性,光镜和电镜下观察脑组织、脑细胞和血脑屏障病理学改变。结果超声联合微泡组和热疗组血脑屏障通透性均显著高于对照组(P<0.05),电镜下微血管内皮紧密连接呈开放状态。结论超声照射联合微泡和全身热疗均可开放血脑屏障。Objective To compare the permeability of blood-brain barrier (BBB) by two different opening methods, ultrasound combined with microbubble destruction and hyperthermia opening in rats. Methods Fourty male Sprague- Dawley (SD) rats were randomly divided into 3 groups. The rats of group A (n=15) were sonicated by ultrasound( 1MHz, 1.2 W/cm )for 3 min after microbubbles injected through femoral vein. Rats of groups B (n=15) were exposed to whole body hyperthermia (WBH) in a heated container maintained at 36℃ for 3 hours. Rats of control group (group C) (n=10) got saline injection into femoral vein. Brain water content was determined by dry-wet weight method, the BBB permeability was measured by Evans blue assay and the state of the interendothelilal tight junction of capillaries was observed with transmission electron microscope (TEM). Results Compared with the normal control, BBB permeability increased and interendothehlal tight junction of capillaries opened, but the abnormalities of nucleo-membrane of neurons was seen in groups B. Conclusion Ultrasound combined with microbubble destruction and hyperthermia could also open BBB.
