摘 要:目的观察醋酸泼尼松对大鼠单位骨中骨矿、骨羟脯氨酸及对大鼠股骨密度和生物力学的影响,研究骨质量的改变与骨生物力学特性改变的联系。方法将20只SSD大鼠,随机分为正常对照组和激素组,每组10只。前者给蒸馏水5ml(/kg.d)灌胃,后者给醋酸泼尼松混悬液3.5mg(/5ml.kg.d)灌胃,88d后心脏抽血处死大鼠。检测单位骨中骨矿及骨羟脯氨酸含量;骨密度和骨生物力学检测。结果醋酸泼尼松抑制大鼠体重的增长,使单位骨中钙及羟脯氨酸含量减少。激素组股骨全段骨密度减小5.18%,股骨远端1cm处骨密度减小7.86%。醋酸泼尼松致股骨力学性能中的最大载荷减小10.78%,弹性载荷减小22.53%,弯曲能量减小24.94%,刚度系数减小18.03%。结论长期应用醋酸泼尼松可致骨基质和矿物质下降,骨密度降低,从而降低了骨的生物力学特性。Objective To investigate the effects of glucocorticoid on bone mineral and bone oxyproline hydroxypreline per osseous tissue in glucoeorticoid-induced osteoporosis rats. To investigate the effects of glucocorticoid on bone mineral density and bio-mechanical property of thighbone in the same model. To hnk the changes of bone quality with biomechanical property. Methods Twenty 5 SD rats were equally divided randomly into control group(CON) given distilled water 5ml/kg/d and model group (GC)given prednisone acetate at dose of 3.5mg/kg/d by intragastric administration. All rats were sacrificed by cardiac puncture under sodium pentobarbital anesthesia 88 days after experiment. Analysis kof bone mineral and and bone oxyproline hydroxyprohne of the left ulna digestive juice and anlaysis of bone mineral density analysis of the left femur were performed in fresh .Bio-mechanics analysis of the right femur was performed in fresh, Results The body weight of model group rats decreased significantly. GC decreased the bone mineraland and bone oxyprehne hydroxyproline per osseous tissue .GC decreased the bone mineral density of femur and the bone mineral density of distal femur. Compared with the control gourp, the maximum load,elastic load,bent energy and stiffness coefficient of rats in the model group were decreased.. Conclusion The decrease of bone matrix, mineral and bone mineral density in glucocorticoid-induced osteoporesis rats was decreased bio-mechanical property.
