摘  要:目的:观察辛伐他汀对3T3-L1脂肪细胞中Apelin mRNA表达影响,为探讨Apelin在心力衰竭发病中的作用以及他汀类药物抗炎作用提供依据。方法:用辛伐他汀溶液刺激诱导分化成熟的3T3-L1脂肪细胞,辛伐他汀浓度分别为0、1、10μmol/L,作用时间设为24h和48h。提取脂肪细胞总RNA,运用实时荧光定量聚合酶链反应技术测定Apelin mRNA表达量的变化。结果:经辛伐他汀干预后3T3-L1脂肪细胞Apelin mRNA表达降低,高浓度的辛伐他汀对Apelin mRNA的表达有显著的抑制作用(P<0.05)。在相同药物浓度下,不同作用时间各组之间差异无显著性(P>0.05)。结论:辛伐他汀可使3T3-L1脂肪细胞中Apelin mRNA表达减少,并具有药物浓度依赖性。这可能部分解释了辛伐他汀的抗炎作用。Objective: To investigate the effect of simvastatin on Apelin mRNA expression in 3T3-L1 adipocytes,so as to know the role of Apelin in heart failure.Methods:The differentiated 3T3-L1 adipocytes were incubated with 0,1,10μmol/L fenofibrate for 24 or 48 hours,then total cellular RNA was extracted, and the expression of Apelin mRNA was detected by Real time Quantitative PCR. Results: Following the treatment of simvastatin, Apelin mRNA expression in 3T3-L1 adipocytes was significantly degraded as the treatment concentration increased (P 〈0.05).Apelin mRNA expression was significantly inhibited by high dose simvastatin .But in same drug concerntrates,there was no significant change in the different action times(P 〉0.05). Conclusion: In 3T3-L1 adipocytes, simvastatin decreases the expression ofApelin mRNA and positively correlates with the drug concentration,whith may partly explain the anti-inflammation effect of simvastatin.
