内耳缺血豚鼠模型内源性硫化氢检测及意义

摘  要:目的:观察内耳缺血豚鼠模型血清内源性硫化氢水平变化并探讨其机制及意义。方法:随机将24只红目豚鼠分为对照组、假手术组及模型组,模型组,采用小脑前下动脉FeCl_3诱导血栓形成制备耳蜗缺血豚鼠模型,造模1小时后,采用激光多普勒血流测量耳蜗血流量,测量实验前后豚鼠听性脑干反应(ABR)域移,采用敏感硫电极法测量血清内源性硫化氢变化。结果:对照组耳蜗血流量变化值0.73%,假手术组组耳蜗血流量减少4.24%,模型组组耳蜗血流量减少37.3%,对照组ABR阈移值0.84%,假手术组ABR阈移值0.92%,模型组ABR阈移值42.1%,对照组血清内源性硫化氢4.1 μmol/L,假手术组血清内源性硫化氢4.0μmol/L,模型组血清内源性硫化氢7.2μmol/L,同对照组及假手术组相比,模型组耳蜗血流量明显减少,ABR阈移值增大,内源性H2S水平升高。结论:内耳缺血造成耳蜗血流量减少,引起听力损伤,H2S升高可能是对抗内耳缺血,保护听力的机制之一。Objective:To observe the change of the serum lever of endogenous hydrogen sulfide in Guinea Pigs,then to investigat its Significance and reason.Methods:The twenty four guinea pigs were randomly divided into three groups: control group,sham operated group and model groups.Thrombus formation upon the anterior inferior cerebral artery induced by FeCl3 was used to develop the cochlear ischemia model in guinea pigs.The cochlear blood flow (CBF)was monitored continuously with tascr Doppler flowmeter, auditory brainstem response(ABR) was monitored before and one hour ate making model,and the serum lever of endogenous hydrogen sulfide was analysis with sensitive electrode in sulphur.Results:The chang of CBF was 0.73% in control group, 4.24% in sham operated group and 37.3% in model groups. The chang of ABR was 0.84% in control group, 0.92% in sham operated group and 42.1% in model groups. The serum lever of endogenous hydrogen sulfidc is 4.1μmol/L in control group, 4.0μmol/L in sham operated group and 7.2μmol/L in model groups.In model groups,the CBF was decreasing, the chang of ABR and serum lever of endogenous hydrogen was increasing.Conclusion:Auris interna ischemia cause decreasing of CBFand hearing toss, the increasing of endogenous hydrogen sulfide lever in serum perhaps is a counteract to auris interna ischemia and hearing loss.
