摘  要:目的建立并完善一种新的体外循环DNA(cDNA)检测方法,并探讨cDNA检测在鼻咽癌(nasopharyr-geal carcinoma,NPC)患者诊断中的应用价值。方法对196例经病理证实的NPC患者以微量基因组抽提试剂盒抽提血浆DNA,以荧光定量Real-time PCR测定含量,并以健康体检者125例作为对照组。结果临床分期各组与治疗前相比,P均<0.05,可见有明显差异;不论治疗前后,NPC患者各组与正常对照组相比,均有明显差异,P均<0.05;cDNA含量与临床分期的关系:根据Ct值的逐渐减少,可见含量水平随着临床分期增加呈增高的趋势,但组间比较并无统计学意义(P>0.05)。结论利用Real-time PCR法测定NPC患者cDNA,对临床NPC的诊断、疗效判断有较大的价值。Objective To quantify the circulating DNA in plasma of patients with nasopharyngeal carcinoma (NPC) and evaluate its potential clinical value. Methods Plasma DNA was extracted by commercial "genomic DNA extraction kit" in 196 patients with NPC and 125 healthy individuals as control and detected by Real-time PCR method. Results There was significant difference in mean circulating plasma DNA levels between NPC patients and healthy individuals (P 〈0. 05), and significant difference between untreated patients and post-treated patients in difference clinical stage (P〈 0. 05). The level of circulating DNA increased gradually with the process of clinical stage. However, there was no significant difference between patient groups in different clinical stages (P〉0. 05). Conclusion Circulating plasma DNA detected by Real-time PCR has potential clinical value in diagnosis and therapeutic effect evaluation of NPC patients.
