摘 要:为了分析BUCHI K-370全自动定氮仪相关参数即蒸汽力度,蒸馏时间的设置使之最适合生物制品中蛋白质含量的测定。分别选用蒸汽力度90%、80%,蒸馏时间190s、170s配对进行硫酸铵回收试验同时对每次回收试验后氨的残留量进行分析比较。再将选出的测定参数对人血白蛋白国家标准品中的蛋白质进行测定。采用蒸汽力度90%,蒸馏时间190s时硫酸铵回收率为100.08%,RSD最小,为0.28%,回收试验后氨残留量最小;且测得标准品的蛋白质含量与真实值对比无显著差异。结果表明采用蒸汽力度90%,蒸馏时间190s最适合生物制品中蛋白含量的测定。To analyze the influence of process control setting such as distillation time and steam power on the protein content detection for biological products. Setting the paramenters , the steam power was 90% or 80% , the distillation time was 190s or 170s. The ammonium sulfate recovery test was carried out and the ammonia residual content was determined after finishing the recovery test each time in amanner that two kinds of parameters were matched each other. The national standard protein of human serum albumin was determined under the given conditions of selected parameters. The tested resuits showed that the ammonium sulfate recovery rate is 100.08%, and RSD is 0.28%, the ammonia residual content is lowest when the distillation time is 190s and the steam power is 90%. The values between the standard protein content and real testing content have no statistical significant difference. The selected parameters { 190s and 90% } are suitable for the detection of protein content in biological products.
