摘 要:目的:建立测定墨旱莲中齐墩果酸的含量的高效液相色谱法。方法:采用Phenomene×C18柱(4.6mm×250mm,5μm),流动相为乙腈-0.1%磷酸水溶液(90:10),检测波长210nm,流速1.0ml/min,柱温为30℃,进样量为20μl。结果:齐墩果酸在0.569-3.414μg内峰面积与其质量浓度有良好的线性关系(r=0.9999);平均加样回收率为98.40%(RSD=2.44%)。结论:该方法简便、快速、准确,可作为墨旱莲质量控制的一个有效方法。Objective: To develop a method of RP-HPLC for the quantitative analysis of oleanolic acid in Herba Ecliptae. Methods: Oleanolic acid was separated on Phenomenex C18 (4.6 mm×250 mm, 5 μm) using a mixture of acetonitrile and 0.1% phosphoric acid solution(90:10) as the mobile phase and detected at 210 nm; The flow rate was 1.0 ml/min and 20 μl. sample solution was injected every time. Results: There was good liner relationship between the peak area and concentration at ranges of 0.569-3.414μg(r=0.999 9) for oleanolic acid; the average recovery rate of oleanolic acid was 98.40% ( RSD=-2.44% ) . Conclusion: The established method was rapid and simple with good accuracy and reproducibility for the determination of oleanolic acid ,the method was suitable for the quality control of Herba Ecliptae.
