扶正抗癌方对大鼠移植性肝癌的实验研究及分析

摘　　要：目的:复制大鼠移植性肝癌模型,探讨扶正抗癌方对大鼠肝癌的疗效及机制。方法:采用Walker256癌肉瘤接种于SD大鼠肝脏,制作移植肝癌模型,分为正常组、假手术组、模型组、扶正抗癌方低、中、高剂量组。记录体质量等生命体征;实验1、2、3w分别检测血清TB、DB、UB;称取瘤重、测量瘤块长短径;取瘤组织、瘤旁组织行病理观察;记录肿瘤转移情况。结果:扶正抗癌方各剂量组大鼠生命体征好于模型组,术后10d模型组大鼠体质量增长加速,可能与腹水产生有关,术后18d高剂量组体质量低于模型组(P<0.05);各剂量中药组胆红素明显低于模型组(P<0.05或P<0.01);扶正抗癌方可抑制肿瘤的生长,其中高剂量组肿瘤质量抑制率高于低剂量组(P<0.05);病理结果示扶正抗癌方可促进癌细胞坏死,抑制其向周围浸润;各剂量组转移评分低于模型组(P<0.05)。结论:扶正抗癌方具有改善移植性肝癌大鼠生存状态、降低胆红素、促进癌细胞坏死、抑制肿瘤生长转移的作用,其中高剂量组效果最突出。Objective： To establish implanted hepatic cancer models in SD rats, and observe the effects and mechanism of Anti-Cancer Herbal Immune Booster on the implanted hepatoma models. METHODS： Walker256 carcinoma was transplanted to the liver of SD rats and the rats were treated with low-dose, medium-dose and high-dose Anti-Cancer Herbal Immune Booster respectively. Their vital signs were observed; serum total bilirubin（TB）, direct bilirubin（DB） and unconjugated bilirubin （UB） were tested; weight and size of liver tumor block were measured, observations of pathological changes in tumor and tumor adjacent tissues were made with hematoxylin-eosin sectioning （HE）; and tumor metastasis were measured at week 1, week 2 and week 3. Results： Compared with the model group, the Anti-Cancer Herbal Immune Booster groups showed evident effects on the vital signs of tested rats. 10 days after operation, the body weight of rats in the model group increased sharply because of ascetics, the body weight of rats in the high-dose group was lighter than that in model group by day 18 after operation （P〈0.05）. All the Anti-Cancer Herbal Immune Booster groups down-regulated the serum bilirubin （P〈0.05 or P 〈 0.01）. The tumor volume and quality decreased, the tumor quality inhibition rate in the high-dose group was higher than that of the low-dose group （P〈0.05）. Furthermore, all the Anti-Cancer Herbal Immune Booster groups demonstrated obvious inhibition of tumor invasion and metastasis in comparison with the model group （P〈0.05）. All Anti-Cancer Herbal Immune Booster groups had significantly higher rate of cancer cell apoptosis, and lower growth rate of tumor cells. Conclusion： Anti-Cancer Herbal Immune Booster, especially high-dose, can improve living quality, down-regulate the levels of serum bilirubin, promote apoptosis of tumor cells and prevent tumor invasion and metastasis.
