摘 要:目的建立反相高效液相色谱法(RP-HPLC)测定虫草制剂中腺苷和尿苷含量的方法。方法采用水浴温浸和加热回流的方法,制备供试品溶液。色谱柱为HypersilC18柱(200mm×4.6mm)流动相为pH6.5磷酸盐缓冲液∶甲醇(85∶15),检测波长为260nm。结果方法的平均加样回收率:腺苷为98.9%,RSD为1.4%(n=6);尿苷为97.9%,RSD为1.5%(n=5),腺苷在23.2~928.0mg/L范围内线性关系良好,尿苷在16.0~640.0mg/L范围内线性关系良好。结论此方法简便、准确,重现性好,可作为判定虫草制剂质量的一种方法。Objective To establish a reversed phase high-performance liquid chromatography (RP-HPLC) method for determining the content of adenosine and uridine in cordyceps preparation. Methods the reagent solution was prepared by water bath digeratur and heating backstream. The chromatographic column was Hypersil C18 column (200 mm×4.6 mm) with mobile phase being pH 6.0 phosphate buffer salinemethanol (85:15), and wavelength being 260 nm. Results The average recovery rate of adenosine was 98.9% with RSD of 1.4 %(n=6) and that of uridine was 97.9% with RSD of 1.5 %(n=5). Adenosine showed a good linear rela tionship in the ranges of 23.2-928.0 mg/L, and uridine in the ranges of 16.0-640.0 mg/L. Conclusion The method is simple, accurate and good reproducibility, it can be used to determine the quality of cordyceps preparation.
